HPLC法测定忍冬果实中绿原酸的含量
Determination of Chlorogenic Acid in Lonicera Japonica Fruit by HPLC
【摘要】 目的:建立以高效液相色谱法测定忍冬果实中绿原酸含量的方法。方法:色谱柱为Kromasil C18反相柱(150mm×3.8mm,5μm),流动相为乙腈-0.05%磷酸水溶液=12:88,检测波长为326nm,流速为0.8mL.min-1。结果:峰面积与浓度在30~150μg.mL-1范围内呈良好线性关系,线性回归方程为Y=21370X-303866(r=0.999),平均加样回收率为98.80%,RSD=1.21%(n=9),样品中的绿原酸含量为0.856%。结论:本方法简便、准确、重现性好,可用于忍冬果实中的绿原酸的含量测定。
【Abstract】 Objective: To determine the content of chlorogenic acid in Lonicera japonica fruit by HPLC.Methods:Chlorogenic acid was separated on Kromasil C18(150mm×3.8mm,5μm) with mobile phase consisted of acetonitrile-0.05% phosphoric acid aqueous solution =12:88 at a flow rate of 0.8mL·min-1.The detective wavelength was set at 326nm.Results:A good linear relationship was achieved over the concentration range of 30~150μg·mL-1 for chlorogenic acid with an average recovery of 98.80%,RSD=1.21%(n=9).The equation of linear regression was Y=21370X-303866(r=0.999),the contents of chlorogenic acid in Lonicera japonica fruit was 0.856%.Conclusion:The method was simple,accurate and sensitive,and it can be used for the determination of Chlorogenic acid in Lonicera japonica fruit.
