【摘要】 目的:研究中药温化蠲痹方对胶原诱导性关节炎(CIA)大鼠外周血TNF-α、IL-1β的影响,探讨温化蠲痹方治疗CIA的作用机制。方法:选用健康雄性Wistar大鼠,随机分为造模组、正常对照组(10只,简称正常组),造模组采用牛Ⅱ型胶原乳剂于大鼠尾根部注射,建立CIA模型。再把造模组随机分为模型组(10只)、MTX组(10只)、温化蠲痹方组(10只,简称中药组)。正常组、模型组、中药组按22.9g/kg剂量分别灌胃生理盐水、温化蠲痹方,均每日1次,MTX组按0.78mg/kg剂量灌胃MTX混悬液,每周1次。给药30天后,运用ELISA法检测大鼠外周血TNF-α、IL-1β水平。结果:与模型组比较,治疗后中药组和MTX组大鼠关节炎指数(AI)评分明显降低(P<0.01);与治疗前比较,中药组和MTX组大鼠AI评分均明显降低(P<0.01)。与正常组比较,模型组大鼠外周血TNF-α、IL-1水平明显升高(P<0.01);与模型组比较,中药组和MTX组大鼠外周血TNF-α、IL-1水平明显降低(P<0.01),两组之间比较差异无显著性(P>0.05)。结论:CIA大鼠外周血TNF-α、IL-1水平升高,降低CIA大鼠外周血TNF-α、IL-1水平可能是中药温化蠲痹方治疗CIA的作用机制之一。更多还原

【Abstract】 Objective:To study the effects on TNF-α and IL-1βin the peripheral blood by Wenhua Juanbi recipe in the rats with Collagen-Inducing Arthritis (CIA).Methods:Healthy Wistar rats were randomly divided into making model group and control group(n=10).Rats of making model group were injected 0.5mL BCⅡ(type Ⅱcollegan of bovine,BCⅡ) emulsion in the tail to establish CIA model.The succeeded CIA rats were randomly divided into three groups,those were model group(n=10),Wenhua Juanbi recipe group(n=10,Chinese heres group for short),MTX group(n=10).Rats of control group,model group and Chinese heres group were given Physiological saline and Wenhua Juanbi recipe respectively by stomach rearing according to 22.9g/kg,rats of MTX group were given MTX liquor according to 0.78mg/kg in the same way.After treatment 30 days,the levels of TNF-α and IL-1β in the peripheral blood were measured by ELISA.Results:AI score of Chinese heres group and MTX group after treatment was lower than that of model group(P<0.01).AI score of those lower than before treatment(P<0.01).The level of TNF-α and IL-1β were significantly higher in peripheral blood of the rats in the model group than that in the control group,the level of TNF-α and IL-1βwere lower in both treated group after treatment than that in the model guoup(P<0.01).Conclusion:The level of TNF-α and IL-1β in peripheral blood increase in CIA rats.It’s one of possible mechenism of treating CIA that Wenhua Juanbi recipe decrease the level of TNF-α and IL-1β in peripheral blood.更多还原
