丹参酮ⅡA对CAM血管生成影响的实验研究

【摘要】 目的探讨丹参酮ⅡA(TsⅡA)对鸡胚绒毛尿囊膜(CAM)模型血管生成作用的影响。方法将鸡胚随机分为生理盐水组、溶媒对照组、bFGF组、TsⅡA0.005mol/L组、TsⅡA0.010mmol/L组、TsⅡA0.015mmol/L组,检测尿囊膜的血管增生程度。结果①各组浓度的TsⅡA均具有较明显的促血管新生作用,与生理盐水组和溶媒对照组相比,差异有统计学意义(P<0.01或P<0.05);TsⅡA0.010mol/L组和TsⅡA0.015mol/L组与bFGF组相比,差异无统计学意义(P>0.05);②具有一定的剂量依赖关系:TsⅡA0.010mol/L组与TsⅡA0.005mol/L组相比,差异有统计学意义(P<0.05),但TsⅡA0.010mol/L组与TsⅡA0.015mol/L组相比,差异无统计学意义(P>0.05)。结论活血化瘀中药单体——TsⅡA可促进CAM的血管增生,具有一定的促血管新生作用。

【Abstract】 Objective To investigate the angiogenenic effects of Tanshinone ⅡA(TsⅡA)on the chick embryo chorioallantoic membrane(CAM).Methods 42 chick embryo(CE)were randomly divided into 6 groups:7 CE in group A were treated with normal saline as negative controls;7 CE in group B were treated with menstruum;7 CE in group C were treated with basic fibroblast growth factor(bFGF)as positive controls;21 CE in group D,E and F were treated with different dose of TsⅡA respectively.After 3 days of incubation,the first and second grade vessels of CAM were counted respectively.Results The proliferation of vessels in experimental groups was more obvious than that in physiological saline group and menstruum group(P<0.01 or P<0.05),and had a certain dose dependent relationship.The proliferation of vessels in TsⅡA 0.010 mol/L group was more obvious than that in TsⅡA 0.005 mol/L group(P<0.05),but difference between TsⅡA 0.015 mol/L group and TsⅡA 0.010 mol/L group was no significant(P>0.05).Conclusion TsⅡA can promote angiogenesis of the chick embryo chorioallantoic membrane.
