运动干预对胰岛素抵抗大鼠血清纤溶酶原激活物抑制剂-1水平的影响
Effect of exercise on the content of serum plasminogen activator inhibitor type-1 in insulin resistant rats
【摘要】 目的观察高脂膳食诱导的胰岛素抵抗(IR)大鼠血清纤溶酶原激活物抑制剂-1(PAI-1)水平的变化以及运动干预对血清PAI-1水平的影响,以初步探讨非药物疗法―运动改善胰岛素抵抗可能的作用机制。方法雄性Wistar大鼠(4周龄,体重70~80 g)随机分为基础饲料组(NC,n=20)和高脂膳食组(HF,n=35)。HF组经特殊高脂膳食喂养10周,建立胰岛素抵抗大鼠动物模型,同时NC组以基础饲料喂养。10周后HF组再随机分为高脂膳食运动组(HFE,n=15)和高脂膳食非运动组(HFNE,n=15),HFE组游泳运动干预4周。以正常血糖–高血浆胰岛素钳夹技术观察高脂膳食喂养大鼠运动前后胰岛素敏感性的变化。酶联免疫吸附法测血清PAI-1水平。提取大鼠肠系膜和附睾周围脂肪代表内脏脂肪,称重。结果以SPSS 16.0统计软件分析。结果葡萄糖钳夹实验结果:10周后HF组葡萄糖输注率(GIR60-120)明显低于NC组(P<0.01),14周后HFE组GIR60-120明显高于HFNE组(P<0.01),HFE组经运动后,GIR60-120明显升高,但仍低于NC组(P<0.01)。与NC组相比,HF组大鼠血清PAI-1水平显著升高(P<0.01),且与内脏脂肪重量成正相关(r=0.656,P<0.05)。运动后HFE组血清PAI-1水平与HFNE组比较显著降低(P<0.01)。结论高脂膳食可构建胰岛素抵抗大鼠模型,其机制与内脏脂肪增加有关。高脂膳食大鼠PAI-1水平升高并与内脏脂肪含量相关。一定负荷量运动后内脏脂肪减少,胰岛素抵抗改善,PAI-1水平降低,提示PAI-1可能参与胰岛素抵抗的发生和发展。
【Abstract】 【Objective】The changes of serum plasminogen activator inhibitor type-1(PAI-1) and the effect of swimming exercise to the content of serum PAI-1 were observed in the high-fat diet-induced insulin resistance rats,in order to explore the possible mechanisms by which the no-drug therapy-exercise improves the insulin resistance.【Methods】Male Wistar rats(4 weeks old,70-80 grams)were randomly assigned to normal control group(group NC) and high-fat-diet feed group(group HF).10 weeks later,group HF was randomly assigned to high-fat-diet feed with exercise group(group HFE) and high-fat-diet feed without exercise group(HFNE),then the HFE groups were arranged to swim for 4 weeks after insulin resistant rats model was established by high-fat diet feeding.The changes of insulin sensitivity after exercise were assessed by euglycemic hyperinsulinemic clamp technique.The serum PAI-1 was measured by ELISA method.The mesentery fat tissue and paraepididymal adipose tissue were weighed at last.SPSS 16.0 was used to analyze all the data.【Results】The glucose infusion rate60-120(GIR60-120) in group HF was significantly lower than that of group NC(P <0.01).The GIR60-120 in group HFE was significantly higher than that of group HFNE(P <0.01),and lower in group HFE than that of group NC(P <0.01).The serum PAI-1 in HF group increased significantly compared with NC group(P <0.01).The serum PAI-1 correlated positively with visceral fat mass(r =0.656,P <0.05).Compared with HFNE group,the serumsPAI-1 decreased significantly in group HFE(P <0.01) after 4 weeks of exercise.【Conclusions】High-fat-diet feed could induce insulin resistance rat’s model.The mechanism of insulin resistance was relevant to elevated visceral fat mass.The mean level of the serum PAI-1 increased in insulin resistant rats,and the elevated PAI-1 level might be correlated with the visceral fat mass.A certain loading dose exercise could diminish the visceral fat mass and the content of serum PAI-1 and improved insulin sensitivity,which indicated that PAI-1 might be involved in the development and the progress of insulin resistance.
