大鼠脂肪间充质干细胞复合羟基磷灰石成骨分化的实验研究

【摘要】 目的脂肪间充质干细胞可与羟基磷灰石复合向成骨细胞分化,检测两种羟基磷灰石复合细胞成骨能力的差别。方法通过扫描电镜观察脂肪间充质干细胞与普通羟基磷灰石和纳米级羟基磷灰石复合情况。检测细胞与载体复合后,表达碱性磷酸酶和骨钙素的情况。结果在将AMSCs细胞种植于两种羟基磷灰石载体表面,向成骨方向诱导4周后,通过扫描电镜观察,羟基磷灰石载体表面有细胞存在。在两种不同的羟基磷灰石(HA)ALP的表达差异有显著性。骨钙素的检测发现,在纳米级羟基磷灰石上的表达要高于普通级羟基磷灰石。结论①脂肪间充质干细胞(AMSCs)可在载体上复合生长。②AMSCs与HA载体复合后,可表达成骨细胞的特征。③诱导成骨后,在纳米级羟基磷灰石上复合表达碱性磷酸酶的量高于普通级羟基磷灰石。

【Abstract】 【Objective】 To observe the bone induction capacity of adipose mesenchymal stem cells(AMSCs) in recombination with hydroxyapatite scaffold and to test the difference in bone induction by two types of hydroxyapatite in recombination with stem cells.【Methods】 By scanning electron microscope,the bone induction capacity of adipose mesenchymal stem cells in recombination with normal hydroxyapatite and nanohydroxyapatite was observed.The expression of alkaline phosphatase(ALP) and BGP was tested.【Results】 Adipose mesenchymal stem cells grew on the surface of two types of hydroxyapatite.After bone induction for 4 weeks,scanning electron microscopy confirmed that the cells existed on the surface of hydroxyapatite.The ALP was(9.782±2.085) u/100 mL on the normal hydroxyapatite,(11.771±3.091) u/100 mL on the nanohydroxyapatite,there was significant difference between the two difference hydroxyapatite groups(P =0.041).BGP expression was higher in the nanohydroxyapatite group than in the normal hydroxyapatite(P =0.012).【Conclusions】(1)AMSCs can recombine with scaffold and grow.(2)After AMSCs recombined with hydroxyapatite,the cells can express the characteristics of the osteoblasts.(3)After bone induction,the expression of ALP on the nanohydroxyapatite is higher than on the normal hydroxyapatite.
