酒精对雌鼠血清泌乳素水平及垂体湿重的影响
Effects of alcohol on plasma prolactin level and pituitary wet weights of female rats
【摘要】 目的探讨酒精对雌鼠血清泌乳素水平及垂体湿重的影响。方法①40只SD同龄成年雌鼠随机分为4组,每组10只,其中3组分别予10%、20%和30%酒精2mL灌胃4周和8周(各5只),对照组予等容积和时间蒸馏水灌胃;②30只4、6和8周龄的SD雌鼠(各10只)随机分为酒精组和对照组(各5只),分别予以20%酒精或等容积蒸馏水灌胃4周。各组大鼠处死后均用放射免疫法检测血清泌乳素水平,并称垂体湿重。结果①30%酒精组灌胃4周时,血清泌乳素水平和垂体湿重明显高于其他3组(P<0.05),灌胃8周时反较4周时显著下降(P<0.05);②10%和20%酒精组灌胃8周后泌乳素水平和垂体湿重较4周时显著升高(P<0.05),且明显高于同期30%酒精组和对照组(P<0.05);③4周龄鼠酒精灌胃后泌乳素水平和垂体湿重显著高于对照组(P<0.05),6周龄和8周龄鼠与对照组相比差异无显著性(P>0.05)。结论高浓度酒精可在一定时间内使血清泌乳素水平升高,但是长期作用反而使得其值下降。长期低浓度酒精摄取可引起血清泌乳素的显著性增高。幼年期摄入酒精更易诱导血清泌乳素升高。酒精可能通过刺激垂体增生改变血泌乳素水平。
【Abstract】 【Objective】To observe the effects of alcohol on plasma prolactin level and pituitary wet weights in female rats.【Methods】① Forty grown-up female SD rats at the same age were randomly divided into three alcoholic groups and one control group with ten in each group.The alcoholic groups involved administration of alcohol for 4(5 rats) and 8 weeks(5 rats) using a liquid ingestion containing 10%,20%,30% ethanol(v/v).The control group was pair-fed with distilled water.②Thirty female SD rats being four,six and eight weeks old were enrolled into 3 age groups(10 in each),each age group was subdivided into alcohol group and control group with 5 in each.Three alcohol groups were administrated with a liquid ingestion containing 20% ethanol(v/v) and the control groups were pair-fed with distilled water for 4 weeks.Prolactin levels in plasma were detected using radioimmunoassay and pituitary wet weights were measured.【Results】 ① 30% ethanol treatment significantly increased the plasma prolactin levels and pituitary wet weights after four weeks alcohol consumption compared to the remaining three groups(P <0.05);however,after continued treatment for 4 weeks both the plasma prolactin levels and pituitary wet weights were decreased(P <0.05).②Compared to the 4 week treatment,10% and 20% ethanol significantly increased the plasma prolactin levels and pituitary wet weights after 8 weeks alcohol consumption(P <0.05) and the measurements were much higher than the remaining two groups(P <0.05).③Only the 4 week-old rats showed significantly higher plasma prolactin levels and pituitary wet weights than those of control group(P <0.05).【Conclusion】Higher alcohol concentrations could increase the plasma prolactin level within a relative short time and the long-term effect is reverse.Long-term lower alcohol concentrations could gradually elevate the plasma prolactin level.The plasma prolactin level could be more easily increased under alcoholconsumption in juvenile stage.The results suggest that ethanol causes hyperprolactinemia by stimulating the hyperplasia of lactotropes in the anterior pituitary gland.
