甲醛对雌性大鼠卵巢颗粒细胞的影响

【摘要】 目的研究大鼠染毒后甲醛对大鼠阴道脱落细胞、卵巢颗粒细胞的显微及超微结构、卵巢颗粒细胞凋亡率的影响。方法将40只SD雌性大鼠随机分为正常组和3个不同浓度甲醛染毒组,腹腔注射甲醛,剂量分别为20.0、2.0和0.2mg/kg,每天1次,连续14d,每天观察阴道脱落细胞变化,14d后处死所有大鼠取卵巢组织,应用苏木精-伊红染色光镜下观察雌性大鼠卵巢病理组织变化,应用电镜技术观察雌性大鼠颗粒细胞超微结构的变化,应用TUNEL染色观察卵巢颗粒细胞凋亡的情况。结果甲醛染毒后,大鼠的阴道脱落细胞周期性变化被扰乱,HE染色后通过光镜观察,甲醛可造成雌性大鼠卵巢的病理组织结构的变化,其病理变化的程度与甲醛呈剂量依赖性;电镜检查发现甲醛可破坏卵巢颗粒细胞的细胞核及细胞器,且与甲醛呈剂量依赖性;TUNEL染色观察到甲醛染毒组大鼠卵巢颗粒细胞的凋亡率随甲醛剂量增加而增加。结论甲醛能够影响大鼠卵巢颗粒细胞的结构,诱导颗粒细胞凋亡,从而影响卵巢的性激素分泌功能。

【Abstract】 ObjectiveTo explore the effects of formaldehyde on the histopathology structure and ultrastructure of vaginal exfoliative cells and ovarian granular cells,and the apoptosis rate of ovarian granular cells in female rats.MethodsHealthy female rats were intraperitoneally injected with different doses of formaldehyde(20.0,2.0,0.2 mg/kg) for 14 consecutive days.Their exfoliative cytoscopy of vagina was done every morning.These rats were killed on the 14th day.Morphological changes of ovary cells were observed by light microscopy and ultrastructure of granular cells were observed by electron microscope.The apoptosis rate of granular cells was determined by terminal deoxynucleotidyl-transferase-mediated dUTP nick-end labeling(TUNEL) staining.ResultsThe periodical change of the vaginal exfoliative cells was disturbed after treatment with formaldehyde.By light microscopy,pathological structure changes in ovarian of female rats increased with dose.By electron microscope,the changes of granular cell ultrastructure increased with dose too.Formaldehyde could induce the apoptosis of granular cells in rats in a dose-dependent manner.ConclusionFormaldehyde has adverse effects on structure of granular cells of the rat ovary,and can cause the apoptosis of granular cells so as to affect the ovarian excretion of hormones.
