链脲佐菌素不同方法诱导SD大鼠糖尿病肾病模型的研究
Induce rat model of diabetic nephropathy by streptozotocin
【摘要】 目的探讨不同方法建立糖尿病肾病(DN)模型的差异,以确定建立糖尿病肾病大鼠模型的最佳方案。方法清洁级健康雌性SD大鼠40只,随机分为4组,每组10只。A组大鼠单侧肾脏切除后腹腔注射链脲佐菌素(STZ)(55mg/kg),B组大鼠单侧肾脏结扎后腹腔注射STZ(55mg/kg),C组单纯腹腔注射STZ(55mg/kg),D组为假手术对照组(注射等量的缓冲液)。大鼠造模后从第1周开始每两周称体重,取血测血糖(Glu)、血肌酐(Scr)、总胆固醇(Chol)和甘油三酯(TG),收集尿测24h尿蛋白(24Upro)、尿肌酐(Ucr),并计算内生肌酐清除率(Ccr)。11周后,处死大鼠,取肾脏称重并进行肾脏病理。结果 A、B两组出现Chol、TG异常增高以及Ccr升高的现象均比C组早。A、B组肾脏超微结构检查系膜细胞增生、基质增多、肾小球基底膜增厚,病理改变比C组更加严重,也更典型。模型组的存活率为C>B>A,成模周期为C>B=A。结论单侧肾脏结扎后再腹腔注射55mg/kg体重STZ可以用来作为建立糖尿病肾病大鼠模型的理想方案。
【Abstract】 【Objective】To investigate the differences of several ways in diabetic nephropathy(DN) rat model induced by streptozotocin(STZ) and to establish the preferred protocol.【Method】Forty SD rats were randomly divided into 4 groups with ten rats each.Group A was to receive intraperitoneal STZ after unilateral nephritic excision,group B was to receive intraperitoneal STZ after unilateral nephritic ligation,group C was to receive intraperitoneal STZ,group D(control group) was to receive buffer solution.The body weight(BW),glucose(Glu),serum creatinine(Scr),cholesterol(Chol),triglyceride(TG) and 24-urine protein(24Upro),urine crea(Ucr) were detected fortnightly and creatinine clearance rate(Ccr) was calculated.After 11 weeks,the rats were killed and then kidney were weighed and taken for pathological examination.【Result】Abnormal increase of Chol,TG and Ccr of the groups A and B was earlier than group C.Ultrastructural study of the kidney showed that proliferation of mesangial cells,increase of mesangial matrix and thickening of glomerular basement membrane of the groups A and B were more worse and representative than the group C.The percents of survival rats were lower and the periods of model making were less.【Conclusion】STZ(55 mg/kg) by i.p after unilateral nephritic ligation is the plan of choice for inducing rat model of diabetic nephropathy.
