广西产牡蛎多糖的制备和抗肿瘤活性初步研究

【摘要】 目的改进制备工艺,分离纯化广西北部湾牡蛎得到牡蛎多糖,并探索其抗肿瘤活性。方法通过匀浆、动物蛋白酶酶解、醇沉及透析等方法,分离获得牡蛎多糖粗品,以广西高发肿瘤鼻咽癌细胞和血管内皮细胞为研究对象,采用MTT法观察牡蛎多糖对细胞生长的作用,HE染色法观察牡蛎多糖对细胞形态学的改变。结果制备得到牡蛎多糖粗品,牡蛎多糖随着浓度的增加,对鼻咽癌细胞和血管内皮细胞的生长抑制率也增加;光镜下观察,细胞体积变小,核浆变少,核固缩,细胞质浓缩,细胞数目减少。结论在一定浓度下,从牡蛎中分离出的牡蛎多糖对鼻咽癌细胞CNE-1和血管内皮细胞的生长增殖具有抑制作用。

【Abstract】 【Objectives】To improve the preparation technology to separate and purify ploysaccharides from the Oyster of Guangxi Province and investigate its antineoplastic activity.【Methods】To get crude product of Oyster Ploysaccharides by means of homogenate,animal albumen proteolysis,depositing in alcohol,dialysis and separation.To observe the effect of Oyster Ploysaccharides on the growth of human cells by means of MTT through the study of Guangxi Provice’s high-incidence nasopharyngeal carcinoma cells and vascular endothelial cells.To observe the effect of Oyster Ploysaccharides on the change of morphocytology by means of HE staining method.【Results】Oyster Ploysaccharides was isolated from Oyster of Guangxi Province.Oyster ploysaccharides was found to have a growing inhibiting effect on nasopharyngeal carcinoma cells and vascular endothelial cell with its growing consistency.Under a microscope,the cells showed morphological changes significantly such as the cell nucleus shrinking,syrup diminishing,karyopyknosis occurring,cytoplasm concentrating and the number of cells reducing.【Conclusion】Ploysaccharides from oyster have a growing inhibiting effect on CNE-1 cells and the multiplicative growth of human vascular endothelial cells at a certain consistency.
