组织工程骨BCBB/BMP/bFGF的成血管作用观察

【摘要】 目的观察吸附有骨形态发生蛋白(BMP)及碱性成纤维细胞生长因子(bFGF)的双相陶瓷生物骨(BCBB)植入兔股部肌袋内的成血管过程,评价bFGF的再血管化作用。方法成年新西兰大白兔60只,随机分为4组,每组15只。A组:股部肌袋内植入BCBB/BMP/bFGF;B组:植入BCBB/bFGF;C组:植入BCBB/BMP;D组:植入BCBB。分别于术后4、8和12周取材,每个时间点每组取材5例标本,进行组织学大体观察、组织切片免疫组化血管染色并进行血管计数和血管面积图像分析。结果组织学大体观察,术后各时间点A组所取标本较B组、C组和D组色泽红润,与周围组织连接紧密。血管计数和血管面积,术后各时间点A组血管数和血管面积均大于B组、C组和D组,B组大于C组和D组,且差异有显著性(P<0.05)。结论BCBB是生长因子的良好吸附载体,适宜新生血管长入。吸附有bFGF及BMP的BCBB具有较强的再血管化能力,BMP能够促进bFGF的再血管化作用。

【Abstract】 【Objective】To observe the pathological changes after BMP/bFGF-adsorbing BCBB(biphasic ceramic biologic bone) being filled into the muscle bag of the rabbit thigh,and evaluate the revascularization effect of bFGF.【Methods】60 rabbits were randomly divided into four groups,which implanted with BCBB/BMP/bFGF(group A),with BCBB/bFGF(group B),with BCBB/BMP(group C) and with BCBB(group D).The specimens were harvested separately at the end of 4,8 and 12 weeks after operation.5 samples were taken off at each interval in every group.A series of examinations were carried out including of gross histology,immunohistochemical vessel staining and vessel image analysis.【Result】 Gross histology,the samples of group A were red and compact join with around tis-sues.Vessel count and vessel surface area,the vessel count and vessel surface area of group A was higher than group B,group C and group D at the 4th,8th and 12th week,and group B was higher than group C and group D,too,and that the differences were obvious(P<0.05).【Conclusion】 BCBB,a kind of fine carrier of growth factors,is suitable for new vessel growth.AACB/BMP/bFGF composite has a strong activity of revascularization effect,and the revascularization effect of bFGF was promoted by BMP.
