壳多糖对增殖的血管平滑肌细胞[Ca2+]i和[H+]i的影响

【摘要】 目的探讨壳多糖对血管平滑肌细胞[Ca2+]i和[H+]i的影响。方法采用荧光探针标记法观察小牛血清对血管平滑肌细胞[Ca2+]i和[H+]i的影响,以及壳多糖对其的干预作用。结果当NBS刺激血管平滑肌细胞后[Ca2+]i浓度和细胞内pH均明显增加;而壳多糖与NBS合用时,胞内[Ca2+]i浓度和pH降低明显降低。结论壳多糖抑制小牛血清诱导的血管平滑肌细胞[Ca2+]i浓度增加和pH上升,说明壳多糖抑制血管平滑肌细胞增殖可能与抑制Ca2+动员有关。

【Abstract】 【Objective】To study the effects of chitosan on [Ca2+]i and [H+]i of vascular smooth muscle cells (VSMC).【Methods】By means of fluorescence probe marking method,we observed the effect of newborn bull serum (NBS,20%) on VSMC [Ca2+]i and [H+]i,and intervention effect of chitosan on theirs.【Result】The [Ca2+]i concentration and pH all increased in cultured VSMC after NBS exposure. These effects were inhibited by chitosan (2000 μg/L).【Conclusion】Chitosan might inhibit Ca2+ mobilization,through which [Ca2+]i concentration and pH of VSMC were inhibited.
