实时荧光定量PCR在副溶血性弧菌食物中毒检测中的应用探讨

【摘要】 目的:应用荧光定量PCR方法快速筛检副溶血性弧菌等食物中毒病原菌,以提高检测速度,与GB培养法比较,探讨两法的符合性和应用价值。方法:采用高度特异、敏感的实时荧光PCR作为初筛方法,用GB方法进行细菌分离培养,用ATB对分离到的病原菌进行鉴定。结果:21份食物中毒病人大便标本实时荧光定量PCR检出副溶血性弧菌特异性DNA核酸阳性18份,GB细菌培养检出18株副溶血性弧菌,两法的阳性符合率一致,检出率均为85.71%。结论:能用实时荧光定量PCR检测食物中毒中的病原菌。与GB培养法相比明显缩短检测周期,报告检验结果及时率提高,达到快速、及时的目的,在食物中毒事件中能更及时查明病因,对临床治疗、流行病学调查和食物中毒处置发挥积极作用。

【Abstract】 Objective:To use the method of real-time Quantitative PCR to accelerate detection of the pathogenic bacteria in the food poisoning caused by Vibrio parahemolyticus to study this method′s coincidence and application value comparing with the GB cultrure method.Methods:Real-time quantitative PCR was applied as the highly specific and sensitive method of preliminary screening.Bacteria was separated by GB method,and then identified by ATB.Results:18 of 21 samples were detected for specific DNA of Vibrio parahemolyticus using the method of real-time quantitative PCR,and the same number of strains were detected by the GB method.Both of the methods have the same detection ratio 85.71%.Conclusion:The method of real-time quantitative PCR is well capable of detecting the pathogenic bactaria causing food poisoning,and accelerating the detecting process comparing with the GB method.It can confirm the cause of bacterial food poisoning in time and contributes to clinic treatment,epidemic survey and treatment of food poisoning.
