PrP105-132在IL-6、IL-8介导下对大鼠原代大脑皮质细胞的影响
Effect of PrP105-132 and IL-6,IL-8 on rat pimary cortical neuron culture
【摘要】 目的探讨朊蛋白病中大脑皮质细胞死亡与朊蛋白、IL-6、IL-8的关系。方法大鼠原代皮质细胞分为对照组、PrP组、IL-6+IL-8组、PrP+IL-6+IL-8组,培养48 h后收集细胞,进行细胞形态学观察、细胞凋亡率及细胞生存率检测。结果 PrP组、PrP+IL-6+IL-8组细胞数目以及细胞生存率均较对照组、IL-6+IL-8组减少,而细胞凋亡率升高(P<0.01)。PrP+IL-6+IL-8组细胞凋亡率明显高于PrP组(P<0.01)、细胞生存率明显低于PrP组(P<0.05)。结论 PrP105-132肽段对大脑原代皮质细胞有毒性作用,能够引起神经细胞凋亡;在IL-6、IL-8存在条件下PrP105-132对大脑原代皮质细胞的毒性作用增强。
【Abstract】 Objective To explore the relationship of cortical neuron death with PrP105-132,interleukin-6 and interleukin-8.Methods The rat pimary corical neuron culture were divided into four groups:the control group,PrP group,IL-6+IL-8 group and PrP+IL-6+IL-8 group.After 48 hours,the cells were collected.The cell morphology changes,apoptosis rate and survival rate were compared.Results The cell counts decreased in PrP group and PrP+IL-6+IL-8 group.The cell apoptosis rate of PrP+IL-6+IL-8 group was significantly higher than that of PrP group(P<0.01).The cell survival rate was significantly lower than that of PrP group(P<0.05).Conclusion PrP105-132 has the toxic effects on primary cortical neuron culture,can result in cell apoptosis.IL-6 and IL-8 may enchance this neurotoxic effects of PrP105-132.
