阿魏酸钠对氧化型低密度脂蛋白诱导的主动脉平滑肌细胞增殖的抑制作用研究

【摘要】 目的观察阿魏酸钠(SF)对氧化型低密度脂蛋白(ox-LDL)诱导大鼠胸主动脉平滑细胞增殖的抑制作用。方法组织块法培养,取大鼠胸主动脉平滑肌细胞进行原代培养,传代后将细胞分为五组,其中ox-LDL组加入100mg/Lox-LDL,SF组在加入25、50、100 mg/ml的SF预处理2 h后,再加入100 mg/Lox-LDL,对照组SMC不加任何干预因素,用MTT法及流式细胞术分析各组细胞的增殖能力及细胞增殖周期。结果原代培养的大鼠胸主动脉SMC多呈长梭形或三角形,传代后形态变化不大,至少可连续培养10-12代;ox-LDL组细胞增殖能力明显高于对照组(P<0.01);不同浓度的SF组细胞增殖能力明显低于ox-LDL组,与ox-LDL组比较,差异具有显著性(P<0.01,P<0.05);与对照组比较,ox-LDL组G0/G1细胞数明显减少,S期和G2/M期细胞数则明显增多,差异具有显著性(P<0.01);与ox-LDL组比较,SF组G0/G1细胞数明显增多,S期和G2/M期细胞数则明显减少(P<0.05,P<0.01),该作用随SF剂量的增加有增强趋势。结论 SF可抑制ox-LDL诱导的大鼠胸主动脉SMC的增殖作用,从而实现其抗动脉粥样硬化作用。
【Abstract】 Objective To discuss the inhibition effects of sodium ferulate(SF) to over-production of smooth muscle cell(SMC) by ox-LDL induced.Methods Patch-attaching method for culture of arterial SMC were adapted,after passaged the SMCs were divided into 5 groups,in ox-LDL group 100 mg/L ox-LDL was added,in SF groups 25,50,100 mg/ml SF were added variedly and 2 h befor 100 mg/L ox-LDL added,only normal culture liquid were added in control group.For all the groups the MTT and flow cytometry were applied to evaluation the production of SMCs.Results The primary SMCs look like long spin or triangle and can be passaged at least 10-12 generation.The production of ox-LDL SMCs were higher significantly than the control group(P<0.01),and compared with ox-LDL group,SF groups increased significantly(P<0.01,P<0.05).Compared with control group,the number of SMCs in ox-LDL group were decreased in G0/G1 phase and increased in S and G2/M phase,the difference is signifiant(P<0.01).Compared with ox-LDLgroup,the number of SMCs in SF group were increased in G0/G1 phase and decreased in S and G2/M phase,the difference is signifiant(P<0.01).Conclusion SF can inhibited the over-production effects of SMC induced by ox-LDL,witch may be helpful for the treatment for AS.
