嘌呤核苷酸补偿对海洛因依赖大鼠嘌呤核苷酸代谢相关酶基因表达的影响
Effects of purine nucleotides compensation on gene expression of enzymes related to purine nucleotides metabolism in heroin dependent rat
【摘要】 目的探讨外源性补偿嘌呤核苷酸对海洛因依赖大鼠嘌呤核苷酸代谢酶的影响。方法将50只雄性Wis-tar大鼠随机分为对照组、海洛因组、海洛因+AMP+GMP组、海洛因+AMP组、海洛因+GMP组。采用real-timePCR的方法检测大鼠脑皮质中腺苷脱氨酶(ADA)、黄嘌呤氧化酶(XO)、次黄嘌呤-鸟嘌呤磷酸核糖转移酶(HGPRT)、腺嘌呤磷酸核糖转移酶(APRT)和腺苷激酶(AK)mRNA的表达水平。结果海洛因组ADA、XO的mRNA比对照组明显升高(P<0.05),而海洛因+AMP+GMP组、海洛因+AMP组、海洛因+GMP组与海洛因组相比明显降低(P<0.05)。海洛因组HGPRT、APRT和AK的mRNA比对照组明显降低(P<0.05),而海洛因+AMP+GMP组、海洛因+AMP组、海洛因+GMP组与海洛因组相比有不同程度升高,以HGPRT的升高程度尤为显著(P<0.05)。海洛因+AMP+GMP组、海洛因+AMP组、海洛因+GMP组的各项指标差异无显著性。结论海洛因在大鼠基因水平上促进嘌呤核苷酸的分解代谢,降低合成代谢,而补偿嘌呤核苷酸可以抑制或拮抗海洛因的上述作用。
【Abstract】 Objective To explore the effect of exogenous purine nucleotides on gene expression of enzymes related to purine nucleotides metabolism in heroin dependent rat.Methods 50 male Wistar rats were randomly divided into control group,heroin administration group,heroin plus AMP and GMP group,heroin plus AMP group and heroin plus GMP group.Real-time PCR were used to examine the relative amount of ADA,GDA,XO,APRT,HGPRT and AK.Results The mRNA of the ADA and XO genes in the brain tissue were increased significantly than that in the control group(P<0.05) and the up-regulation of the expression of the ADA and XO genes induced by heroin treatment could be reversed by purine nucleotides.The mRNA of the AK,APRT and HGPRT genes in the brain tissue were decreased significantly than that in the control group(P<0.05) and the down-regulation of the expression of the AK,APRT and HGPRT genes induced by heroin treatment could be reversed by purine nucleotides (P<0.05).Conclusion Purine nucleotides compensation could ameliorate the enhancement of purine nucleotides catabolism and the degression of purine nucleotides anabolism in heroin dependent rats.
