1株长春地区羊传染性脓疱病毒的分离鉴定及F1L基因的克隆与序列分析

【摘要】 利用MDBK细胞从长春郊区一养羊户送检的出现典型"口疮"症状的病羊唇部痂皮中分离获得1株病毒,通过电镜观察、理化学测定、PCR检测、动物回归试验、间接免疫荧光(IFA)及病理组织学观察等方法对该株病毒进行了系统鉴定,证实该分离株为羊传染性脓疱病毒(Orfvirus,ORFV),命名为ORFV-cc。参照GenBank中ORFVF1L基因的核苷酸序列,设计合成1对特异性引物,成功地对ORFV-cc株F1L基因进行克隆、测序,并推导出其氨基酸序列。序列分析表明,获得的ORFV-cc分离株F1L基因片段长987bp,编码328个氨基酸,与ORFV-NZ2,ORFV-7,ORFV-20,ORFV-C2,ORFV-mi-90,ORFV-Torino株核苷酸序列同源性分别为97.5%,98.3%,97.9%,98.5%,98.4%,97.5%;氨基酸序列同源性分别为97.2%,97.2%,97.5%,98.8%,98.8%,97.9%。系统发育进化树结果表明,ORFV-cc分离株与参考毒株ORFV-C2和ORFV-mi-90的亲缘关系较近,表明不同地区分离毒株的F1L基因差异不大。

【Abstract】 An Orf virus strain(ORFV-cc) was successfully isolated from scab materials of lambs with clinical sore mouth symptom by passaging in MDBK cells.The virus was identified as Orf virus based on morphology,physico-chemical property,PCR,IFA,animal regression test and pathohistology observation.A pair of primers derived from the F1L gene of Orf virus(ORFV) was designed according to the published sequences in GenBank.The F1L gene was cloned and sequenced,then the amino acids sequence of F1L protein was deduced.The F1L gene contained 987 bp,which encoded a protein of 328 amino acids.The F1L gene of the ORFV-cc strain shared a nucleotide identity of 97.5%,98.3%,97.9%,98.5%,98.4%,97.5% and an amino acid identity of 97.2%,97.2%,97.5%,98.8%,98.8%,97.9% with those of strain ORFV-NZ2,ORFV-7,ORFV-20,ORFV-C2,ORFV-mi-90,ORFV-Torino,respectively.The results of the phylogenetic tree analysis indicated that there was a close genetic relationship between ORFV-cc strain and ORFV-C2,ORFV-mi-90.These indicated the F1L gene of different strains was not obvious the difference.
