他汀类药物对EAE小鼠RhoA表达的影响

【摘要】 目的探讨他汀类药物能否抑制小鼠实验性自身免疫性脑脊髓炎(EAE)模型RhoA蛋白表达及其对小鼠发病情况的影响。方法随机将雌性C57BL/6小鼠分为EAE组、他汀治疗组和佐剂组3组。EAE组和他汀治疗组用髓鞘少突胶质细胞糖蛋白多肽(MOG35-55)免疫小鼠建立EAE模型,佐剂组以生理盐水替代MOG35-55同样方法注射;他汀治疗组自免疫后第3天始按体质量10 nag/(kg·d)予阿托伐他汀灌胃。观察3组小鼠发病情况及病理变化并测定脑组织中RhoA蛋白表达。结果与佐剂组相比,EAE组小鼠脑组织中RhoA蛋白表达明显增多(P<0.01)。他汀治疗组脑组织RhoA蛋白表达及临床神经功能评分均较EAE组下降(P<0.01)。结论 EAE小鼠RhoA蛋白表达上调。他汀类药物可能通过抑制RhoA蛋白表达治疗EAE。

【Abstract】 Objective To investigate the effects of statin on the RhoA protein expression and the incidence of disease in the experimental autoimmune encephalomyelitis (EAE) mice. Methods All female C57BL/6 mice were randomly divided into 3 groups: the EAE group, the statin group and the adjuvant group. Mice in EAE and statin groups were immunized with myelin oligodendrocyte glycoprotein peptides 35-55 (MOG35-55), the adjuvant group was injected at the same time, but instead of MOG35-55 with normal saline. Atorvastatin was intragastrically injected in the dosage of 10 mg/ (kg·d) in the statin group on the 3rd day post-immunization. The clinical features were observed and the histopathology was detected by HE staining. And RhoA protein expression of brain were detected. Results The activation of RboA protein in EAE group was obviously increased compared with those of adjuvant group (P<0.01). Brain RhoA protein expression in the statin group was decreased compared with the EAE group, as did the clinical scores (P<0.01). Conclusions The expression of RhoA protein was upregulated significantly in EAE mice. The mechanism underlying of the effect on EAE may be correlated with the downregnlation ability on RhoA protein expression and activation.
