鹿茸柱前衍生化高效液相指纹图谱研究
Study on Pre-column Derivatization HPLC Fingerprint of Cornu Cervi Pantotrichum
【摘要】 目的:建立鹿茸的柱前衍生化高效液相指纹图谱,为科学评价及有效控制其质量提供有效可靠的方法。方法:采用2,4-二硝基氟苯(DNFB)为衍生化试剂对鹿茸药材进行处理。高效液相色谱条件为PhenomenexLuna 5u C18(2)100R(4.6 mm×250 mm,5μm)色谱柱,甲醇-醋酸钠缓冲液梯度洗脱,流速1.0 mL·min-1,柱温20℃,检测波长360 nm。结果:在本实验条件下测定10批鹿茸药材,其色谱指纹图谱可反映鹿茸所含的17种氨基酸成分,当中所含的10种必需氨基酸的含量与市售价格有较好的一致性。结论:该方法精密度、稳定性、重复性好,为鹿茸的质量控制提供了思路和导向。
【Abstract】 Objective:To establish a pre-column derivatization method for HPLC fingerprint analysis of Cornu Cervi Pantotrichum.Method:Cornu Cervi Pantotrichum was derivated by 2,4-dinitrofluorobenzene.HPLC analysis was performed on Column Phenomenex Luna 5u C18(2) 100R(4.6 mm ×250 mm,5 μm),with a mixture of methanol and acetate buffer as mobile phase in gradient mode.The flow rate was 1.0 mL.min-1,wavelength was set at 360 nm,and column temperature was at 20 ℃.Result: Seventeen kinds of amino acids in Cornu Cervi Pantotrichum were detected,of which 10 kinds of essential amino acids showed a concordance with the content and market price.Conclusion: Good precision,stability and repetition were showed.The method provides ideas and guide in the quality control of the valuable Cornu Cervi Pantotrichum.
