山楂中齐墩果酸和熊果酸的测定
Determination of Oleanic Acid and Ursonic Acid in Crude Hawthorn Berry by HPLC
【摘要】 目的:建立测定山楂果中齐墩果酸和熊果酸含量的高效液相色谱法。方法:采用C18色谱柱,甲醇-水-磷酸-三乙胺(87∶13∶0.2∶0.04)为流动相,流速0.9mL·min-1,35℃,检测波长210nm。结果:齐墩果酸和熊果酸的分离度>1.4;方法学考察符合液相色谱法的要求;山楂药材中齐墩果酸含量>0.5mg·g-1;熊果酸含量>1.9mg·g-1。结论:该法简便、灵敏、分离度高、专属性好,适用于山楂中齐墩果酸和熊果酸的含量测定。
【Abstract】 Objective:To establish an HPLC method for determination the content of Oleanic acid and Ursonic acid in crude hawthorn berry. Method: The HPLC analysis was carried out through C18 column with detection wavelength set at 210 nm and the temparature set at 35 ℃ . The mobile phase was composed of MeOHH2O-H3PO4-TEA (87∶ 13∶ 0. 2∶ 0. 04) at a flow rate of 0. 9 mL·min -1. Result:The separation of Oleanic acid and Ursonic acid was 1. 4; the methodologic validation conformed to requirement of liquid chromato-graphy. The content of Oleanic acid and Ursonic acid in crude hawthorn berry was more than 0. 5 mg·g -1 and 1. 9 mg·g -1, respectively. Conclusion:The HPLC method can be used in the routine analysis of Oleanic acid and Ursonic acid in crude hawthorn berry.
