苦茶的急性毒性与神经药理活性初步研究

【摘要】 苦茶Camellia kucha是我国特有的一类野生茶树资源,其化学成分上不同于其他茶树资源的特殊性在于富含1,3,7,9-四甲基尿酸(苦茶碱)。对苦茶的安全性及其对中枢神经系统的药理作用进行了初步探讨。结果表明,小鼠每kg体质量苦茶急性毒性经口LD50>21.50 g,按毒性分级标准判定属无毒级;苦茶中的苦茶碱对戊巴比妥钠的中枢抑制有协同作用,具有镇静催眠活性,在苦茶中可以拮抗咖啡碱的中枢兴奋作用;当这两种成分含量相当的时候,苦茶没有表现出明显的催眠或兴奋作用;但当苦茶碱的含量是咖啡碱的3倍时,苦茶具有镇静催眠作用。

【Abstract】 Kucha(Camellia kucha) is a special wild tea plant containing a great amount of theacrine(1,3,7,9-tetramethyluric acid) besides caffeine and theobromine.Acute toxicity and sedative/hypnotic activity of Kucha were studied.The result showed that the LD50 of Kucha was more than 21.50 gkg bw,suggested that the water extract of Kucha had no toxicity under our experimental conditions.Theacrine could obviously prolong the sleeping time of mice induced by pentobarbital sodium,while caffeine had the opposite exciting effect.When the contents of these two components were about similar in Kucha,no significant sedative and hypnotic effects or exciting effect was observed.However,when The contents of theacrine was 3 fold higher than that of caffeine,Kucha showed potent sedative and hypnotic properties.
