BDNF对新生鼠缺血缺氧性脑损伤的保护作用的研究
Protective effect of BDNF to newborn rats with hypoxic-ischemic brain damage
【摘要】 目的:研究提前给予脑源性神经生长因子(Brain-derived neurotrophic factor,BDNF)对新生鼠的缺血缺氧性脑损伤(Hypoxic-ischemic brain damage,HIBD)的保护作用。方法:选取7 d龄Wistar大鼠256只,随机分成4组,①假手术组(n=64,A),②模型组(n=64,B),③模型前给药组(n=64,C),④模型后给药组(n=64,D),每组又分为1、3、5、7 d 4个时间点,每个时间点8只,用免疫组织化学和实时荧光定量PCR方法检测脑组织中海马区域的Trk-B和p75的表达水平。结果:模型前注射组和模型后注射组的Trk-B和p75水平分别与对照组和假手术组比较均有显著性差异(P<0.05)。结论:缺血缺氧模型前给予BDNF可保护HIBD引起的脑损伤,其可能机制是通过提高Trk-B的表达水平和降低p75的表达水平来发挥作用的。
【Abstract】 Objective:To study the protective effect of BDNF to newborn rats with hypoxic-ischemic brain damage before the modeling.Methods:Two hundred and fifty-six 7-day-old Wistar rats were randomly divided into four groups: sham operation group(n=64,A);model group(n=64,B);model pre-treated group(n=64,C);model post-treated group(n=64,D),every group are further divided into 1、3、5、 7 d four time points(n=8).The expression of Trk-B and p75 in Hippocampus was examined with immunohisto chemical staining and real-time PCR.Results:Trk-B and p75 levels in model pre-treated group and model post-treated group,had significant differences compaed with model group and sham operation group,P<0.05.Conclusion:BDNF has a protective effect to the brain injury caused by HIBD. Enhancing the expression of Trk-B levels and reduce the expression level of p75 may play a role.
