OPN在口腔扁平苔藓中的表达及作用初步研究

【摘要】 目的:检测骨桥蛋白(Osteopontin,OPN)在口腔扁平苔藓(Orallichen planus,OLP)局部粘膜和外周血中的表达水平,探索其在OLP免疫发病机制中的作用。方法:免疫组化法检测OPN在局部粘膜组织的表达;ELISA检测外周血清OPN和白细胞介素12(Interleukin-12,IL-12)的蛋白浓度;分离正常人外周血单个核细胞(Peripheral blood mononuclear cells,PBMCs),分OPN刺激组和对照组,体外活化72小时,流式细胞学检测淋巴细胞的凋亡率。结果:OPN在OLP患者局部病变粘膜组织的表达较健康对照组明显增强(P<0.01);OPN和IL-12在OLP患者血清中的浓度均明显高于健康对照组(P<0.01),而且二者存在显著正相关(r2=0.6585,P<0.0001);体外活化淋巴细胞的凋亡率,OPN刺激组明显低于对照组(P<0.05)。结论:OPN在OLP局部粘膜和外周血中的表达水平明显升高,并且可以抑制体外活化淋巴细胞的凋亡,提示OPN在OLP的发病发展中可能起着重要的促进作用。

【Abstract】 Objective:To study the expression of Osteopontin(OPN)in local oral mucosal tissue and sera from patients with oral lichen planus(OLP),and to explore the immunologic mechanism of OLP.Methods:Local oral mucosal tissue was examined for OPN by immunohistochemical analysis.Serum OPN and IL-12 levels were measured by enzyme-linked immunoabsorbent assay.PBMCs isolated from peripheral blood of the healthy volunteers were activated in the presence or absence of recombinant human protein OPN (rhOPN) for 72 h.The apoptosis of activated lymphocytes was analyzed by flow cytometer.Results:OPN was upregulated significantly in local oral mucosal tissue from OLP compared with NC (P<0.01).Serum OPN and IL-12 concentration in OLP was significantly higher compared with NC(P<0.01).Moreover,a significant positive correlation (r2=0.658 5,P<0.000 1) was observed between serum OPN and IL-12 concentration in patients with OLP.The percentage of apoptotic cells activated with OPN was significantly lower compared with that of control(P<0.05).Conclusion:OPN was upregulated in OLP and could inhibit apoptosis of activated lymphocytes.OPN may play a key role in the pathogenesis of OLP.
