人参皂甙(Rh2)抗动脉粥样硬化作用实验研究
Experimental Studies on Anti-atherosclerotic Effects and Mechanisms of Ginsenoside Rh2
【摘要】 目的探讨人参皂甙(ginsenoside Rh2,G-Rh2)对动脉粥样硬化大鼠血脂、血清NO、MDA含量、SOD活力的影响.方法将45只Wistar大鼠随机分为对照组、模型组、Rh2组,每组15只.对照组大鼠喂饲普通饲料,模型组及Rh2组大鼠采用高脂饮食加维生素D3(总量7×105U/kg,分别于第1周和第6周腹腔注射)建立大鼠动脉粥样硬化模型.Rh2组大鼠高脂喂养同时1次/d Rh2200 mg/(kg.d)-1灌胃,每周称体质量1次,根据体质量变化调整给药量;对照组、模型组大鼠以生理盐水灌胃.11周后颈动脉采血,检测血脂水平,血清一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平.结果 1)Rh2对血脂的影响:与对照组相比较,模型组大鼠血清TC,LDL-c及AI显著升高(P<0.01);与模型组比较,Rh2给药组TC,LDL-c和AI明显降低,差异显著(分别P<0.01,P<0.01,P<0.05);而高密度脂蛋白、血清三酰甘油水平相比无统计学意义.2)Rh2对血清NO含量的影响:与对照组相比较,模型组大鼠血清NO含量明显降低(P<0.01);与模型组比较,Rh2组NO含量明显升高(P<0.05);药物组与对照组相比无统计学意义.3)Rh2对血清MDA含量与SOD活力的影响:模型组MDA水平与对照组相比明显升高(P<0.01),而SOD活力明显降低(P<0.01).与模型组相比,Rh2对血清SOD活力有明显升高的作用(P<0.05);同时,Rh2具有显著降低MDA水平的作用(P<0.05).Rh2组与对照组相比无统计学意义.结论 Rh2通过降低高脂大鼠血清低密度脂蛋白和总胆固醇水平,提升高脂大鼠血清NO水平、血清SOD活力,降低MDA含量,增强抗脂质过氧化功能和清除氧自由基,稳定细胞膜,保护内皮细胞而发挥抗AS作用.
【Abstract】 Objective To investigate the effects of Rh2 on lipemia,NO,MDA and SOD in the atherosclerosal Rats.MethodExperimental atherosclerosis in rats was produced by feeding with high fat diet and administering vitamin D3(7×105 U/kg) in intraperitoneal injection at the first week and the sixth week respectively.Healthy forty-five male or female Wistar rats,whose body weights range from 180 g to 230 g.The three groups were normal group(feed on normal diet),AS model group(feed on high fat diet) and Rh2 group(feed on high fat diet and Rh2 200 mg/(kg·d).The former two groups were given isotonic sodium chloride instead of Rh2accordingly.The rats of each group were weighed once a week and the dosages were adjusted to the changes of body weight accordingly.After the rats were fed for successive 11 weeks,serum levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),Nitric oxide(NO),Superoxide dismutase(SOD),Malondialdehyde(MDA) were detected respectively.Results1)The effect of Rh2 on blood lipoproteins:The serum level of TC,LDL-c and AI of Model group was significantly higher than that of Control group(P<0.01);Rh2 decreased the serum level of TC,LDL-c,and AI significantly.The difference of each index between Model group and Rh2 group was remarkable(P<0.01,P<0.01,P<0.05)but there was no statistic difference between them in TG and HDL-c.2)The effect of Rh2 on the serum NO level:The serum NO level of Rh2 group was higher than that of model group(P<0.05).The NO level of control group was higher than that of model group(P<0.01);The difference in NO level between Rh2 group and control group was not remarkable statistically.3)The effects of Rh2 on the serum MDA level and SOD activity:MDA level of model group increased obviously compared with that of control group(P<0.01).SOD activity of model group decreased obviously compared with that of control group(P<0.01).Rh2 highly improved the serum SOD activity compared with model group(P<0.05) and highly decreased the serum MDA level compared with model group(P<0.05).The differences in MDA level and SOD activity between Rh2 and control group were not remarkable statistically.ConclusionBy decreasing serum levels of TC,LDL-c,by improving serum levels of NO,by improving activity of SOD and decreasing the level of MDA,Rh2 would protect vascular endothelium and prevent AS in this way.
