黑麦麸皮色素纯化工艺及稳定性研究
Purification and Stability Evaluation of Pigment from Black Wheat Bran
【摘要】 以河东乌麦麸皮为原料,通过大孔吸附树脂的静态试验和动态试验确定纯化黑麦麸皮色素的工艺。试验结果表明:最佳试验吸附树脂确定为HZ801;吸附条件在温度30℃,pH3.0,供试液吸光度为0.8,吸附流速为1 BV/h时,吸附效果较佳;用70%乙醇,pH 1.0,温度30℃,解吸流速为3 BV/h时,洗脱作用较好。色素在丙酮、三氯甲烷、石油醚、正丁醇中不溶,而能溶于乙醇、甲醇、水;可耐短时高温,85℃加热1 h,色素的残余率为93.35%;日光灯处、黑暗处较稳定;在pH值为0.5~3.0时较为稳定;色素对NaC l较稳定。产品得率为0.91%,色价为11.1。树脂纯化后黑麦麸皮色素杂质含量少、纯度高。为黑麦麸皮色素进一步具体的研究和开发提供一定的参考依据。
【Abstract】 Taking Hedong black wheat bran as raw material and through macroporous resin static testing and dynamic testing,the process of preparaing black wheat bran pigment was established.Result:The determined proper experimental adsorption resin is HZ801;the established adsorption conditions for a good adsorption effect are temperature 30 ℃,pH 3.0,test liquid absorbance 0.8,and adsorption flow rate 1 BV/h;the established desorption conditions are 70% ethanol,pH 1.0,temperature 30 ℃,and desorption flow rate 3 BV/h.The pigment is insoluble in acetone,chloroform,petroleum ether,n-butanol,but soluble in ethanol,methanol and water;the pigment can bear short-time high-temperature and the pigment remnant rate is 93.35% after heating for 1h at 85 ℃.The pigment is comparatively stable in fluorescent light and at dark,comparatively stable under the condition of pH 0.5~3.0,and stable to NaCl.The product yield is 0.91% and the product color value is 11.1.The resin purification produces black wheat bran pigment of less impurity and high purity.
