2-甲氧基雌二醇对SKM-1细胞凋亡中bcl-2/bax表达的影响
Effect of 2-methoxyestradiol on cell apoptosis and the expression of bcl-2/bax in SKM-1 cells
【摘要】 目的探讨2-甲氧基雌二醇(2-ME)对骨髓增生异常综合征SKM-1细胞凋亡过程中bcl-2/bax表达的影响。方法 2-ME与SKM-1细胞共同培养,流式细胞术分析细胞凋亡率和细胞周期;RT-PCR技术检测bcl-2和bax mRNA表达。结果 1~8μmol/L 2-ME作用后,SKM-1细胞凋亡率上升呈剂量依赖性;细胞被阻滞于G2/M期;细胞bcl-2 mRNA表达量下调,bax mRNA表达量上调。结论 2-ME诱导SKM-1细胞凋亡与细胞G2/M期阻滞和bcl-2/bax比率下降有关。
【Abstract】 Objective To explore the effect of 2-methoxyestradiol(2-ME) on cell apoptosis and the expression of bcl-2/bax ratio in myelodysplastic syndromes SKM-1 cells.Methods After SKM-1 cells were treated with different concentration of 2-ME for several period of time,the rate of apoptosis and cell cycle status were evaluated by FCM,and the expressions of bcl-2 and bax mRNA were assayed by reverse transcription polymerase chain reaction(RT-PCR).Results Different concentration of 2-ME increased the rate of apoptosis in a time-dependent manner and caused a sustained arrest at G2/M phase.Furthermore,2-ME significantly down-regulated the expression of bcl-2 mRNA(P<0.05) and up-regulated the expression of Bax mRNA(P<0.05).Conclusions 2-ME could induce SKM-1 cell apoptosis by arresting cell cycle at G2/M phase and decrease the ratio of bax/bcl-2.
