黄连解毒汤对MPTP所致帕金森病小鼠的防治作用

【摘要】 目的观察黄连解毒汤(HJD)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)所致C57BL小鼠帕金森病模型的防治作用。方法 C57BL小鼠腹腔注射MPTP30mg·kg-1.d-1,连续5d,诱导小鼠帕金森病模型。观察HJD对小鼠肢体运动(爬杆、悬挂、游泳)功能的影响;HPLC-ECD法检测小鼠脑纹状体内多巴胺(DA)及其代谢产物二羟基苯乙酸(DOPAC)、高香草酸(HVA)的含量;分光光度计法测定小鼠脑组织超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-Px)及丙二醛(MDA)的含量。结果与对照组比较,模型组小鼠肢体运动功能明显降低(P<0.05,P<0.01),纹状体内DA、DOPAC及HVA的含量明显减少(P<0.01),脑组织中SOD及GSH-Px酶活力明显降低,但MDA的含量却显著升高(P<0.01);HJD对小鼠运动功能的减退和脑神经组织的代谢异常有不同程度的调节和改善作用。结论黄连解毒汤对MPTP所致帕金森病小鼠有一定的防治作用。

【Abstract】 Objective To observe the protective effect of Huanglian Jiedu decoction (HJD) on mice with Parkinson’s disease (PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Methods 45 male C57BL mice were evenly randomized into 3 groups:control,MPTP model (subcutaneous injection 30 mg·kg-1·d-1 for 5 days) and HDJ+MPTP groups (17.55 g crude drug/kg). HJD was given for 16 days by2intragastric administration and subcutaneous injection 30 mg/kg for 5 days at the eighth day. Performance of animals in the pole,traction and swimming test were observed at 4 days after the last injection. Levels of dopamine (DA) and its metabolites 〔3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA)〕 in the striatum of animals were measured by high-performance liquid chromatography with an electrochemical detector (HPLC-ECD). The spectrophotometer method was used to examine the levels of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px ) and malondialdehyde (MDA). Results Comparing with the control group,the model group had obvious effect on the behavioral performance of the animals,and the decreased score in the pole,traction and swimming test(P<0.05 or P<0.01)and the striatal levels of DA,DOPAC and HVA were significantly lower(P<0.01),and the activity of SOD and GSH-Px were improved obviously in the brain,but the content of MDA was significantly higher(P<0.05 or P<0.01). Comparing with the model group,HDJ+MPTP group had significantly improved in the different indexes(P<0.05 or P<0.01). Conclusions The preventive medication of HJD shows a protective effect on C57BL mice with PD induced by MPTP.
