Wortmannilactone F产生菌邬氏黄丝曲霉F01Z0195的培养条件优化

【摘要】 本研究通过单因素实验和响应面分析法相结合的方法,以新组织蛋白酶B抑制剂wortmannilactone F的生成量为考察对象,对邬氏黄丝曲霉F01Z0195的培养条件进行优化。确立邬氏黄丝曲霉F01Z0195的培养方式为固态静置发酵培养,培养基为含水的玉米渣,接种量7%,培养周期为14d,最优培养基为40.0g玉米渣和18.3mL含有2.2%DMSO的1/15mol/L磷酸盐缓冲液(自来水配制,pH值为7.46),在此条件下wortmannilactone F生成量为3.90mg/g,与理论值基本一致,与初始培养条件相比提高了341倍。

【Abstract】 In this study,with methods of single-factor experiments and response surface analysis,the cultural conditions of Talaromyces wortmannii F01Z0195 were optimized based on the production of wortmannilactone F,which was a new cathepsin B inhibitor.The best cultural manner was solid-state fermentation for 14 days with 7% inoculation volume,the optimal culture media were composed of 40.0g corn grit and 18.3mL phosphate buffer with 1/15mol/L(prepared with tap water,pH 7.46) containing of 2.2% DMSO.Under the optimal conditions,the production of wortmannilactone F increased to 3.90mg/g,which was 341 times to the initial value,the experimental value was consistent with theoretical value.
