高效液相色谱法测定左旋尤利沙星的含量及有关物质

【摘要】 目的建立测定左旋尤利沙星含量及有关物质的高效液相色谱方法。方法采用DiamonsilC18柱(250mm×4.6mm,5μm),以乙腈-0.2%三乙胺和0.1%庚烷磺酸钠水溶液(用磷酸调节pH2.5)(25:75)为流动相,流速为1.0ml/min,检测波长为275nm,柱温为35℃。结果主峰能与相邻杂质峰达基线分离,左旋尤利沙星的浓度在10~200μg/ml范围内线性关系良好,相关系数为0.9996,最低检测限为0.2ng。结论该法简便、准确,灵敏度高,专属性强,可以用于左旋尤利沙星的含量及有关物质的测定。

【Abstract】 Objective To develop an HPLC method for determination of levoulifloxacin and its related substances.Methods The chromatographic separation of levoulifloxacin was performed on a Diamonsil C18 column with a mobile phase consisting of acetonitrile-0.2%triethylamine and 0.1%sodium heptanesulfonic acid at pH 2.5 adjusted with phosphoric acid(25:75)with a flow rate of 1.0ml/min.The detection was monitored at 275 nm,and the column temperature was maintained at 35℃.Results Levoulifloxacin was completely separated from its related substances.The calibration curve of levoulifloxacin was linear in the range of 10-200μg/ml with the correlation coefficient of 0.9996.The limit of detection was 0.2ng.Conclusions The method is simple,rapid,sensitive and specific,and can be used for determination of levoulifloxacin and its related substances.
