中性粒细胞趋化因子在轻、重症急性胰腺炎大鼠胰腺组织中的表达变化
Expression of cytokine-induced neutrophil chemoattractant in mild or severe acute pancreatitis rats
【摘要】 目的:探讨中性粒细胞趋化因子(cytokine-induced neutrophil chemoattractant,CINC)在轻症急性胰腺炎(mild acute pancreatitis,MAP)向重症急性胰腺炎(severe acute pancreatitis,SAP)演变过程中的作用。方法:分别采用腹腔注射雨蛙肽和胆胰管逆行注射5%牛磺胆酸钠建立大鼠轻症、重症急性胰腺炎模型。雄性SD大鼠随机分为雨蛙肽组(18只)、生理盐水组(18只)、牛磺胆酸钠组(24只)和手术对照组(24只),检测各组不同时间点血清淀粉酶并观察胰腺组织病理学改变,用免疫组织化学法和半定量逆转录-聚合酶链反应(RT-PCR)法检测胰腺组织中CINC蛋白和CINC mRNA表达的变化情况。结果:雨蛙肽组6,12 h胰腺组织中CINC蛋白和CINC mRNA表达明显高于生理盐水组(P<0.05),且表达与胰腺组织的病理积分呈正相关;24 h与生理盐水组比较无明显差异。牛磺胆酸钠组与手术对照组相比,大鼠的血清淀粉酶明显增高、胰腺组织学改变明显;术后1 h胰腺腺泡细胞CINC蛋白和CINC mRNA的表达高于生理盐水组,随着时间的延长表达逐渐增强(P<0.05,P<0.01)。结论:CINC可能在MAP向SAP演变过程中起到重要作用。
【Abstract】 Objective: To investigate the role of cytokine-induced neutrophil chemoattractant(CINC) in the course of exchange from mild acute pancreatitis(MAP) to severe acute pancreatitis(SAP).Methods: Acute pancreatitis model was induced by intraperitoneal injection of cerulein or retrograde infusion of 5% sodium taurocholate into the bili-pancreatic duct in SD rats.SD rats were randomly divided into four groups:cerulein group(eighteen),saline control group(eighteen),sodium taurocholate group(twenty four) and operational control group(twenty four).Serum amylase level and the histological score of pancreas were measured at different time point in every group.Expression of pancreatic CINC proteins and CINC mRNA were determined by immunohistochemistry and semi-quantitative reverse transcriptase polymerase chain reaction(RT-PCR),respectively.Results: Pancreatic CINC proteins and CINC mRNA were increased in cerulein group in comparison with saline control group 6,12 hours following the first injection(P<0.05).Expression of CINC was correlated with the histological score of pancreas.No significant difference was found between cerulein group and saline control group after 24 hours.Serum amylase and the histological score of sodium taurocholate group were significantly higher than those of operational control group.Expression of CINC proteins and CINC mRNA in pancreatic acinar cells in sodium taurocholate group 1 hour after operation were significantly higher than those of operational control group.Expression of CINC were gradually up-regulated(P<0.05,P<0.01).Conclusion: CINC may play an important role in the pathogenesis of AP and in the course of exchange from MAP to SAP.
