弓形虫入侵宿主细胞时SAG3基因表达抑制对β-微管蛋白mRNA水平的影响
The effect of the suppressed expression of SAG3 of Toxoplasma gondii on the mRNA level of β-tubulin during parasite invasion
【摘要】 目的研究表面抗原3(SAG3)基因表达被抑制对弓形虫β-微管蛋白mRNA水平的影响。方法用电穿孔的方法将针对SAG3基因3′-UTR区的dsRNA转入弓形虫速殖子体内,转染后的虫体感染Hela细胞,感染后5min提取总RNA,RT-PCR检测SAG3基因的抑制效率,并分别于感染后5min、30min和1h检测β-微管蛋白mRNA水平。结果实验组SAG3基因表达被抑制,弓形虫转染宿主细胞后5min、30min和1h的β-微管蛋白mRNA水平均降低。结论SAG3影响弓形虫入侵蛋白β-微管蛋白mRNA水平。
【Abstract】 Objective To study the effect of the suppressed expression of SAG3 of Toxoplasma gondii on the level of β-tubulin during parasite invasion.Methods dsRNA targeting 3-UTR of SAG3 was transfected into a T.gondii RH strain by electroporation.Then,the parasites infected Hela cells.Five min after infection,total RNA was extracted,and the inhibition of SAG3 expression was detected by semi-quantitative RT-PCR.The levels of mRNA of β-tubulin were detected by RT-PCR 5 min,30 min,and 1 h after infection.Results The level of mRNA of β-tubulin in the experimental groups was lower than that in the control group.Conclusion Suppressed expression of SAG3 can modulate the expression of β-tubulin of T.gondii.
