HPLC测定不同居群苍术中苍术素醇的含量

【摘要】 目的:应用RP-HPLC方法,对不同居群苍术中苍术素醇的含量进行比较。方法:高效液相色谱法,色谱柱Agi-lent TC-C18(4.6 mm×250 mm,5μm),柱温为30℃,流动相为乙腈-水(49∶51),检测波长为337 nm,流速为1.0 mL.min-1。结果:苍术素醇在9.12×10-2~9.12 mg.L-1(r=0.999 9)呈现良好的线性关系;平均回收率为97.15%,RSD 1.5%(n=5);大别山区苍术各居群样品中苍术素醇含量比江苏茅苍术高出4~6倍。结论:建立了一种准确、简便的苍术质量评价方法;不同居群苍术中的苍术素醇含量与其形态及地理分布有一定的相关性。

【Abstract】 Objective: To establish an RP-HPLC method for determination of atractylodinol in Ateractylodes lancea and compare the contents of atractylodinol in the herbs of different origins.Method: The samples were separated on an Agilent TC-C18(4.6 mm×250 mm,5 μm) column with the mobile phase of acetonitrile-water(49∶51).Flow rate was 1.0 mL·min-1.The detection wave length was set at 337 nm.Column temperature was 30 ℃.Result: The linear range of atractylodinol was 9.12×10-2-9.12 mg·L-1(r=0.999 9),the average recovery was 97.15%,RSD was 1.5%(n=5).The contents of atractylodinol were in the range of 0.268-1.213 mg·g-1 in the samples from different orgins.The contents of atractylodinol in samples growing in Dabieshan mountain were higher than those in Jiangsu province(P<0.001).Conclusion: The established method for determination of atractylodinol is accurate and reliable,which can be used to evaluate the quality of A.lancea,the contents of atractylodinol in the sample was related with its morphological characteristic and geographic orgin.
