吸烟对原癌基因c-fos在骨组织表达的影响

【摘要】 目的探讨原癌基因c-fos在吸烟诱导骨质疏松中的可能作用机制。方法 选取吸烟和不吸烟健康成年志愿者各20名,分为吸烟组和非吸烟组,吸烟组患者为每天吸烟20支以上,烟龄不少于10年;非吸烟组患者为不吸烟或每天吸烟不超过1支。各取约1克髂骨松质骨,提取其总RNA和总蛋白,通过RT-PCR测定c-fos mRNA的表达;通过Western blotting测定Fos蛋白表达;通过免疫沉淀法测定Fos蛋白磷酸化程度。结果吸烟组c-fos mRNA表达高于非吸烟组,差异有统计学意义(P<0.05)。Western blotting检测,吸烟组Fos值高于非吸烟组,差异有统计学意义(P<0.01)。结论 吸烟可以导致人骨组织c-fos基因与Fos蛋白表达增加,提示原癌基因c-fos与吸烟诱导骨质疏松关系密切,可能c-fos表达增高与骨细胞和成骨细胞的凋亡有关。

【Abstract】 Objective To explore the possible functional mechanism of proto-oncogene c-fos in smoking-induced osteoporosis.Methods Twenty healthy adult smoker volunteers and 20 non-smoker volunteers were included in the study.They were divided into smoking group and non-smoking group.People in smoking group were determined by smoking at least 20 cigarettes a day and lasting at least for 10 years.People in non-smoking group were determined by no smoking at all or smoking not more than 1 cigarette a day.One gram of bone tissue in ilium was extracted from each volunteer.Total RNA and total protein were extracted from the samples.The expression of c-fos mRNA and Fos protein were detected using RT-PCR and Western blotting.The degree of Fos protein phosphorylation was detected using immuno-precipitation.Results The c-fos mRNA expression in smoking group was higher than that in non-smoking group,and the difference was statistically significant(P < 0.05).The Western blotting detection showed that Fos protein expression in smoking group was higher than that in non-smoking group,and the difference was statistically significant(P < 0.01).Conclusions Smoking could lead to increased expressions of both c-fos mRNA and Fos protein in human bone tissue.This suggested that proto-oncogene c-fos might be closely related to smoking-induced osteoporosis.The increased expressions of c-fos might be related to the apoptosis of osteocytes and osteoblasts.
