STK33基因在肺癌 乳腺癌 结肠癌组织中表达的研究

【摘要】 目的:探讨STK33基因在肺癌、乳腺癌、结肠癌组织中表达及其与临床分期的关系。方法:选择2007年1月至2009年4月接受手术治疗并有完整临床资料的肺癌、乳腺癌、结肠癌患者174例。采用免疫组织化学染色及HE染色的检测方法,对174例肺癌、乳腺癌、结肠癌组织中STK33蛋白表达进行观察,并分析它们之间的关系。结果:在正常肺组织及肺良性病变组织中均失表达,肺癌组织中表达率为79.7%,肺癌转移淋巴组织中表达率为100%;肺癌及转移淋巴结组织中STK33蛋白表达率显著高于正常肺组织及肺良性病变组织(P<0.01)。肺癌组织中STK33基因表达水平与肺癌肿瘤细胞分化程度、患者淋巴结转移有密切关系(P<0.05)。乳腺癌和结肠癌组织中STK33阳性率与对应的非癌组织之间有显著差异。用Western-Blot分析显示,肺癌组织中STK33的表达水平远高于非癌组织。结论:STK33蛋白高表达强度与恶性肿瘤肺癌、乳腺癌、结肠癌病变的恶性程度呈正相关,随着肿瘤的发展STK33蛋白表达水平呈递增趋势,标志着STK33蛋白的表达增高使肿瘤细胞更具有恶性表型和恶性行为。提示STK33蛋白可能成为早期非小细胞肺癌患者术后评价预后的重要分子标志物,可以作为肺、乳腺、结肠病变诊断的参考指标。

【Abstract】 Objective: To study expression of STK33 protein in lung cancer, mammary gland cancer, and colon cancer tissue and its relation to clinical stage. Methods: Tests were conducted on 174 lung cancer, mammary gland cancer, and colon cancer tissue samples and examined together with additional patient management data from January 2007 to April 2009. Immunohistochemical technique and H&E staining were used to observe expression of STK33 protein in the lung cancer, mammary gland cancer, and colon cancer tissue samples. Results: STK33 lost its expression in 20 benign lesion of lung and normal lungs, the expression of STK33 was 79.7% and 100% in lung cancers (51/64) and lung cancer carcinomatous metastasis lymphoid tissue (16/16). There was a significant difference of expressionin STK33 levels in lung cancer and lymphaden metastasis compared with normal lung tissue and lung benign lesions (P<0.01). The expression status of STK33 in lung cancers was associated with the lung cancer tumor cell differentiation and the lymphaden metastasis (P< 0.05). The expression of STK33 in mammary gland cancers and colon cancers were significantly different to non-cancerous tissue (P<0.05). Western blot analysis demonstrated higher expression of STK33 in lung cancers than in non-cancerous tissue. Conclusion: A high expression of STK33 is related with malignant tumors in lung cancer, mammary gland cancer and colon cancer and can be used as a reference sign for the diagnosis of pathological changes in the lung, mammary gland, and colon. As a tumor develops, the expression level of STK33 tends to increase, thus raised expression of STK33 protein indicates malignant phenotype and malignant behavior. This implies that STK33 protein could become a molecule marker for post-operative evaluation for the prognosis of non-small cell lung cancer patients.
