淫羊藿黄酮治疗骨质疏松性骨折的动物实验研究
Experimental Research of Epimedium-derived-flavonoids for Repairing Osteoporotic Fracture
【摘要】 目的:研究淫羊藿黄酮对动物性骨质疏松性骨折愈合的治疗效能。方法:选取36只4月龄C57雌性小鼠先行腰1椎体骨密度检测后切除双侧卵巢(OVX),3周末再次行腰1椎体骨密度检测,建立右股骨中段骨折克氏针内固定模型,随即分成实验组18只(以淫羊藿黄酮化合物20mg/kg/day、钙剂56mg/kg/day喂药,每日1次)和对照组18只(钙剂56mg/kg/d),于骨折第7天,14天和28天分别处死后使用X线摄片观察骨折端愈合情况、用定量CT获取的骨折段皮质骨密度(cBMD)值及其横截面积(CSA)后,计算两者乘积视为骨力学强度指数(BSICSA),比较骨折端力学强度指数(BSICSA)和骨密度(cBMD)变化并作统计学分析。结果:C57雌性小鼠OVX 3周后,定量CT检测的腰1椎体骨密度下降比较具有统计学意义(P<0.01),X线片显示实验组和对照组相应时点比较,骨折修复的时间明显缩短;经定量CT获取的骨折段第7天,14天和28天皮质骨力学强度指数(BSICSA)、骨密度(cBMD)变化F组别值为15.72(P<0.01)和F组别值为64.05(p<0.05),显示出淫羊藿黄酮组和对照组在骨折修复早、中、晚期骨折端密度、强度的比较差异具有统计学意义,淫羊藿黄酮治疗和时间因素均对骨折修复有影响,其中以淫羊藿黄酮治疗因素最有效,Eta组别>Eta时间*组别>Eta时间;结论:淫羊藿黄酮能有效促进实验性骨质疏松性骨折修复。
【Abstract】 objective:To observe the repairing effect of Epimedium Flavonoids(EF) on the animal osteoporotic fracture.Methods:Thirty-six female 4-month-old C57 mice after being performed bone mass density(BMD) measurement of 1st lumbar vertebrae with quantitative computed tomography(QCT) at pre-and post-resection of bilateral ovarian(OVX),and made the open fracture model of right femur with kirsches wire intramedullary fixation,were randomly divided into two groups: experimental group(fed by EF 20mg/kg and calcium 56mg/kg/day once a day) and control group(fed by calcium 56mg/kg/day once a day).Bone union time,the cortical cross sectional area(CSA) and the cortical BMD were detected by QCT on 7 th,14 nd and 28 th days after been sacrificed respectively.Also,the average of BMD and bone strength index(BSI)(cortical CSA×cortical BMD) around 0.6 cm of fracture site(3 slices) were calculated and statistically analyzed.Results:After three weeks of OVX,the BMD of 1st lumbar vertebrae of the mice were significantly decreased.Compared with the control group,the experimental group’s BSI(Fgroup=15.72,P<0.01) and cortical BMD(Fgroup=64.05,P<0.05) were all of significant statistical difference,the effect of interaction of FE factor was the best(Etagroup>Etagroup*time>Etatime).Conclusion: EF can obviously shorten the repairing time of animal osteoporotic fracture.
