粒细胞集落刺激因子对脑出血大鼠的神经保护作用
Protective effect of granulocyte colony-stimulating factor on intra-cerebral hemorrhage in rats
【摘要】 目的:观察粒细胞集落刺激因子(G-CSF)对脑出血大鼠的神经保护作用并探讨其机制。方法:胶原酶定位注射构建脑出血大鼠模型,然后给予G-CSF腹腔注射连续5d,通过肢体放置实验检测大鼠神经功能恢复情况,并检测给药后出血灶面积的改变,通过免疫组化检测G-CSF促进出血灶周边神经发生的情况,用Hoechest33258荧光染色的方法检测G-CSF的抗凋亡作用。结果:术后给予G-CSF的大鼠肢体置放实验评分明显高于未给药组;给予G-CSF的大鼠出血灶周围可检测到神经干细胞,且出血灶所在大脑半球神经细胞凋亡明显低于未给药组。但给药组与未给药组出血灶面积无明显区别。结论:G-CSF能够促进脑出血大鼠的神经功能的恢复,对脑出血大鼠有神经保护作用,其机制可能涉及其抗凋亡和促进神经发生的功能。
【Abstract】 OBJECTIVE To study the potential neuro-protective effect of G-CSF in a rat model of intra-cerebral hemorrhage(ICH) and the possible mechanisms underlying the effect.METHODS Experimental ICH was induced by the stereotaxic administration of collagenase type Ⅶ injected into caudate nucleus.The recombinant human G-CSF(15 μg·kg-1),dissolved in normal saline,was administered through intra-peritoneal,at 24 h after the ICH induction and daily afterwards for 5 days.The limb placement tests were examined weekly for 4 weeks after intracerebral hemorrhage was induced.Then the expression of nestin was measured by immunohistochemistry,apoptosis was observed by the immunofluorescence staining of Hoechst33258,and the sizes of necrosis area were also measured.RESULTS It was found that intraperitoneal injection of G-CSF for 5 days could efficiently attenuate the ICH-induced neuronal behavioral impairment.G-CSF may stimulate the expression of neural stem cells in the peri-necrosis area of the hemorrhagic hemisphere in the rats and G-CSF had anti-apoptotic effect after ICH.But no differences were found in the sizes of necrosis area between G-CSF-treated group and control.CONCLUSION G-CSF has neuroprotective effect in a rat model of ICH.The mechanism may involve proliferation of neural stem cells and the anti-apoptotic effect of G-CSF.
