GC/MS-NCI法检测头发中四氢大麻酚酸
GC/MS-NCI Determination of 11-nor-△~9-tetrahydrocannabinol-9-carboxylicacid in Human Hair
【摘要】 目的采用GC/MS法对毛发中的△9-四氢大麻酚(THC)及其主要代谢物△9-四氢大麻酚酸(THCA)进行测定。方法气相色谱条件:HP-5MS色谱柱;进样口温度:250℃;柱温:100℃(1min)-30℃.min-1-270℃(3min);载气为氦气,流速为1.0mL.min-1(恒流模式);不分流,进样量:2μL。结果用NCI作为离子源,THCA在0.01~0.2ng.mg-1范围内响应值与样品浓度呈良好的线性关系。采用NCI离子源检测THCA,最低检出限(LOD)为3.0pg.mg-1。用NCI离子源检测TH-CA,高浓度(0.2ng.mg-1)和低浓度(0.02ng.mg-1)时方法日内重复性RSD分别为16.4%,24.8%,高浓度(0.2ng.mg-1)时日间重复性RSD为26.2%。用NCI离子源检测THCA,高浓度(0.2ng.mg-1)和低浓度(0.02ng.mg-1)时方法的回收率分别为82.3%、86.5%。结论本方法简便快速,可用于人体头发中大麻类物质的确证分析。
【Abstract】 Objective To establish a GC/MS method for the analysis of 11-nor-△9-tetrahydrocannabinol-9-carboxylicacid(THCA) in hair of drug abusers. Methods Use the HP-5MS column; the temperature of Injector is 250℃; keep oven temperature 100℃ for 1 min,then increase the temperature to 270℃ at the rate of 30℃·min-1,finally keep the temperature for 3 min. Use the Helium as carrier and keep the speed at 1.0mL·min-1(at constant flow mode). Inject 2μL of sample onto the column in splitless mode. Results The linear correlation of THCA in hair is observed from the concentrations of 0.01~0.2ng·mg-1.The LOD is 3.0pg·mg-1.The RSD values in one day are 16.4% and 24.8% at high concentration (0.2ng·mg-1) and low concentration (0.02ng·mg-1),respectively. The RSD value in different day is 26.2%. The recoveries are 82.3% and 86.5% at high concentration (0.2ng·mg-1) and low concentration (0.02ng·mg-1),respectively. Conclusion The method is fastness and convenience. It provides the evidence information of Cannabis abuse.
