【摘要】 目的探讨VEGF基因转染对VEGF诱导后血管内皮细胞生物学功能的影响,为组织工程血管构建提供血管内皮细胞的可能性。方法应用VEGF体外诱导兔骨髓间充质干细胞定向诱导分化成血管内皮细胞后,再通过脂质体介导的方法将pCDI/VEGF121转染分化后的血管内皮细胞,通过MTT及流式细胞仪检测转染前后细胞的增殖与凋亡能力。结果诱导培养2周后兔骨髓基质细胞转化为血管内皮细胞,VEGF基因转染血管内皮细胞后表达VEGF蛋白,转染后细胞增殖能力增加,凋亡能力下降。结论转染VEGF是促进VEGF诱导血管内皮细胞增殖的有效途径,可望为组织工程血管构建提供血管内皮细胞衬层,为人工血管内皮化开辟一种新的途径。

【Abstract】 Objective To investigate the effect of VEGF gene transfection on the biological function of vascular endothelia cell induced by VEGF and provide the possibility of the vascular endothelia cell for tissue engineering vascular construction.Methods After VEGF induce rabbit mesenchymal stem cells were differentiated into vascular endothelia cell by VEGF in vitro,we detect the ability of cell proliferation and apoptosis by MTT and flow cytometry methods after CDI/VEGF121 were transfected into the cells by liposome-mediated method.Results rMSCs were induced for 14 days by VEGF and transformed into vascular endothelia cells,in which the expression of VEGF was found.and The ability of cell proliferation and apoptosis were increased and decreased respectively.Conclusion Targeting VEGF gene was an effective pathway to promote the vascular endothelia cells proliferation.It could provide the vascular endothelial underlay for tissue engineering construction and develop a new pathway for vascular prosthesis endothelialization.
