白刺总黄酮及单体化合物对过氧化氢损伤内皮细胞的修复作用研究

【摘要】 目的:研究白刺总黄酮及单体化合物对过氧化氢(H2O2)损伤内皮细胞的修复作用及机制。方法:用H2O2诱导血管内皮细胞损伤,以酶标仪检测细胞内一氧化氮(NO)、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)以测定氧化的程度,评价黄酮单体化合物槲皮素、山柰酚、异鼠李素的药理作用。结果:不同浓度的黄酮单体化合物槲皮素、山柰酚、异鼠李素可促进受损的内皮细胞修复,同时提高SOD、NOS、GSH-PX活力和NO水平。结论:白刺总黄酮及单体化合物具有保护血管内皮细胞损伤的作用,其中山柰酚的修复作用最强,其机制与其抗氧化作用有关。

【Abstract】 OBJECTIVE: To study the effects of total flavonoid and monome compound of Nitraria tangutorun on the vascular endothelial cells injury induced by hydrogen peroxide and its mechanism. METHODS:Vascular endothelial cells injury was induced by hydrogen peroxide. The level of nitrogen monoxidum (NO), nitric oxide synthase (NOS), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) in the cells with SN-3001 Microplate Spectrophotometer to evaluate the degree of oxidation. The pharmacological action of quercetin, campherol and isorhamnetin were evaluated. RESULTS: The different concentration of quercetin, campherol and isorhamnetin promote the repair of impaired vascular endothelial cells and the activity of SOD, NOS and GSH-PX activity and the content of NO. CONCLUSION: These results demonstrate that total flavonoid and monome compound of N. tangutorun play the protective action on hydrogen peroxide, especially campherol; its mechanism may be related to antioxidant effect.
