微生物实验室压力蒸汽灭菌质量控制

【摘要】 目的了解微生物实验室压力蒸汽灭菌效果状况,以提高和保证压力蒸汽灭菌质量。方法采用工艺监测和生物指示剂监测方法,对实验室压力蒸汽灭菌质量进行了验证和监控。结果经对所选择的压力蒸汽灭菌器的灭菌工艺验证,装载容量控制在70%条件下,对盛装于500ml三角烧瓶培养基的灭菌工艺确定为121℃20min。用留点温度计进行252次灭菌温度监测,灭菌温度合格率为100%,化学指示剂监测合格率为98.0%。在设定灭菌温度121℃,作用时间20min,用嗜热脂肪芽孢杆菌芽孢进行15次灭菌效果监测,合格率为100%。结论经过工艺验证,所确定的灭菌条件,可确保压力蒸汽灭菌质量。

【Abstract】 Objective To know the the effects of pressure steam sterilization in microbiological laboratory,and to improve and ensure the quality of pressure steam sterilization.Methods Processing monitor and biological indicator monitor were used to monitor the quality of pressure steam sterilization of laboratory.Results When the filling rate was below 70%,121℃ 20 min was chosen as the sterilizing parameter by sterilizing the culture medium in 500 ml flasks.The qualified rate with 252 tests for stationary point thermometer tests was 100% and that for chemical indicator tests was 98.0%.The qualified rate with 15 tests of sterilization efficacy tested by Bacillus stearothermophilus spore was 100%.Conclusion By process testing,the sterilization parameter we chose can ensure the quantity of the pressure steam sterilization.
