抗生长分化因子-9抗体与化疗所致大鼠卵巢损伤的关系的研究

【摘要】 目的:探讨抗生长分化因子-9抗体(Anti-GDF-9)对化疗所致大鼠卵巢损伤是否具有防治作用。方法:将96个大鼠卵巢分为6组:①正常对照组12个;②磷酰胺氮芥(PM)组12个;③生理盐水组12个;④同时加抗体组20个;⑤先加抗体组20个;⑥后加抗体组20个。加入抗体的3组有12个卵巢培养8 d,8个卵巢培养14 d,其余3组均培养8 d,隔天换液;按上述要求取出标本,称湿重,HE染色,计数卵泡总数。结果:培养8 d,正常对照组和同时加抗体组的卵泡总数比较,差异无统计学意义(P>0.05),其余各组两两比较卵泡总数差异均有统计学意义(P<0.05),其中先加抗体组多于正常对照组;培养14 d,加入抗体的3组两两比较卵泡总数差异均有统计学意义,先加抗体组卵泡总数最多。结论:体外实验,在加入PM前或同时加入Anti-GDF-9可能会减轻PM对大鼠卵巢的损伤。

【Abstract】 Objective: To study the effect of Anti-Growth differentiation faotor-9(Anti-GDF-9) against glyciphosphoramide(PM) induced ovarian damage in female rats.Methods: Ninety-six ovaries(forty-eight Wistar rats)were randomly divided into six groups,such as normal control group(n=12),PM group(n=12),saline group(n=12),PM+ Anti-GDF-9 groups(with Anti-GDF-9 administerd simultaneously,before and after PM separately for 24 h,20 each).To culture the ovarian tissue in 24 hole culture plates,one ovary each hole,for 8 days or 14 days(PM+ Anti-GDF-9 groups,8 ovaries each group),replaced culture medium(no PM) every two days for all ovaries.Weighting,HE staining and counting the number of follicles of each phase.Results: There showed significant difference in the number of follices between every two groups(P<0.05) except between the normal control group and the group which administerd Anti-GDF-9 simultaneously with PM at day 8 after culture,and the number of follicles of the group which administerd Anti-GDF-9 before PM was more than the normal control group.There showed significant difference in the number of follices between every two groups of the PM+Anti-GDF-9 groups at day 14 after cultured,and the group which administerd Anti-GDF-9 before PM had the largest number of follicles.Conclusion: Anti-GDF-9 administerd before PM for 24 h and simultaneously with PM can diminished PM induced ovarian damage in ovaries in vitro.
