高脂饮食对大鼠胰岛细胞凋亡的影响
【摘要】 目的:探讨高脂饮食对大鼠胰岛细胞凋亡的影响和机制。方法:20只雄性Wister大鼠,随机分两组,其中对照组给予普通大鼠饲料,实验组给予含混合油脂的高脂饲料。10周后抽血并测定血糖、胰岛素、甘油三酯、胆固醇后取胰腺,并用末端脱氧核苷酸转移酶介导duTP缺口末端标记法(TUNEL)检测大鼠胰导细胞中的细胞凋亡率,观察两组对大鼠胰导细胞凋亡的影响。结果:高脂饮食组较对照组血脂升高,胰岛细胞凋亡率高于对照组。结论:高脂饮食明显影响大鼠血脂水平,高脂饮食使胰岛细胞凋亡率明显增高。
【Abstract】 Objective:To investigate effects of high level of fatty acid on the apoptosis of rat pancreatic islet cells.Methods:twenty 10-weeks-old healthy male Wister rats were assigned at random into two groups.The control group was fed with normal chew and experimental group were fed with high fat diet by adding mixed fat into normal chew.After feeding for 10 weeks,fasting plasma insulin,plasma glucose ,triglyceride and cholesterol were analyzed.We examined islet beta cells by the terminal deoxy nucleotidyl transfer rase biotin-dUTP nick and labeling(TUNEL) in each group,then calculated positive cell number of the apoptotic cells.Results:the high fatty acid intake groups ,results of the proportion of the apoptotic cell of primary rat islet cell remarkably higher than of the control group(P<0.05).Conclusions:Long term high fat diet can leads to hyperlipoidemin High level of free fatty acids may initiate the apoptosis of islet beta cells,Apoptosis is one of the reasons for the lose of islets.
