响应曲面法优化微波辅助提取苹果渣多酚工艺研究

【摘要】 为从苹果渣中提取具有生理活性的多酚物质,在单因素试验的基础上,采用响应曲面法优化微波辅助提取苹果多酚的工艺,建立该工艺的二次多项数学模型,研究微波功率、提取时间、乙醇体积分数和料液比4个因素及其交互作用对提取工艺的影响。试验结果表明,对苹果多酚得率的影响次序是:微波功率﹥料液比﹥提取时间﹥乙醇体积分数;微波辅助提取苹果渣多酚的最佳工艺条件是:微波功率650W、提取时间53s、乙醇体积分数60%、料液比1:20(g/mL),多酚得率达61.8286mg/100g干果渣。

【Abstract】 China is the largest producer of apples in the world.More and more apple pomace is generated by apple juice factories.It has been proven that apple pomace is a rich source of polyphenols,which have a broad spectrum of bioactivities,such as antioxidation,antimicrobe,antitumor,antihypertension,antihyperlipemia,et al.In order to effectively extract polyphenolic compounds from apple pomace,investigation of microwave-assisted extraction was carried out.The whole experiments were carefully designed and optimized with response surface method(RSM).A second order quadratic equation for polyphenols extraction was finally obtained.Factors,such as microwave power,microwave-assisted extraction time,ethanol concentration,ratio of solvent to pomace(g/mL) and their corresponding relationships were investigated.Results showed that the effect order of four factors is as follows:microwave power>ratio of solvent to pomace > microwave-assisted extraction time>ethanol concentration.The optimized extraction conditions were microwave power 650 W,microwave time 53s,ethanol concentration 60%,ratio of solvent to pomace 1:20(g/mL).Under the optimal conditions,the extraction rate of apple polyphenols from apple pomace reached to 61.8286 mg/100 g dry wet.
