替米沙坦对自发性高血压大鼠淋巴细胞电压依赖性钾通道的阻断作用

【摘要】 目的通过全细胞膜片钳技术观察自发性高血压大鼠(SHR)外周血淋巴细胞电压依赖性钾通道(Kv)的电流变化及其替米沙坦对该通道的阻断作用,探讨淋巴细胞Kv在高血压病中的变化及其替米沙坦对该通道的阻断作用。方法将实验动物分成2组,SHR为实验组,正常Wistar大鼠为对照组,每组各10只。使用密度离心法分离SHR和正常血压大鼠外周血淋巴细胞,采用全细胞膜片钳技术记录表达在淋巴细胞上Kv的电流特性,使用不同浓度替米沙坦(10、30、100μmol/L)灌流观察其对SHR淋巴细胞Kv电流的影响。结果 SHR组淋巴细胞Kv峰值电流密度为(119.0±9.6)pA/pF(n=30),对照组为(56.0±8.5)pA/pF(n=40),差异有统计学意义(P<0.05)。替米沙坦呈浓度依赖性阻断SHR淋巴细胞Kv,不同浓度替米沙坦(10、30、100μmol/L)对Kv的阻断率分别为(10.5±3.4)%、(45.8±3.7)%、(81.6±4.2)%。结论 SHR淋巴细胞Kv电流密度增加,替米沙坦呈浓度依赖性阻断Kv,替米沙坦可能通过阻断淋巴细胞上的Kv发挥炎症调节作用。

【Abstract】 Objective To study the change of current of voltage dependent K channel (Kv) in lymphocytes from spontaneously hypertensive rat (SHR) and the effects of telmisartan on Kv. Methods Peripheral blood lymphocyte were collected from 10 spontaneously hypertensive rats and 10 wistar rats. Peripheral blood lymphocytes of SHR and normotensive rats were isolated by density centrifugation. The whole-cell Kv were recorded with patch-clamp technique and current of Kv was observed in response to telmisartan(10,30,100 mol/L). ResultsThe densities of Kv were higher in lymphocytes from SHRs [(119±9.6)pA/pF] than in controls [(56±8.5)pA/pF,P<0.05]. Telmisartan resulted in a concentration dependent inhibition effect on Kv in a range of 10-100 μmol/L of telmisartan,with peak inhibition rate at (81.6±4.2)%. Conclusions The lymphocyte Kv are higher in SHR suggesting involvement Kv in the pathophysiology of hypertension. Telmisartan blocks Kv which could be one of the mechanisms related to its anti-inflammation effects.
