恩诺沙星免疫抗原的制备与鉴定
Synthesis and Identification of Enrofloxacin Antigen
【摘要】 通过N-羟基琥珀酰亚胺活化酯法合成恩诺沙星免疫抗原,用聚丙烯酰胺凝胶电泳、紫外光谱和酶联免疫法分析对恩诺沙星免疫抗原的结构、分子量、免疫原性等特征进行鉴定。经紫外光谱分析恩诺沙星-牛血清白蛋白偶联比为5:1,紫外光谱峰形发生变化并位移。间接酶联免疫法分析证明其免疫兔所获得的抗体与恩诺沙星有良好的特异性,半数抑制浓度为60ng/mL。实验结果表明成功合成了恩诺沙星免疫抗原,并获得了抗恩诺沙星抗体,为ELI SA或TRFI A等免疫方法的进一步研究提供了试验基础。
【Abstract】 The immunogen ENR-BSA(enrofloxacin-bovine serum albumin) were synthesized by the N-Hydroxy-succinimide active ester method.The conjugations were identified by SDS-PAGE,UV and animal immunization test.The coupling molar ratios of the hapten and carrier proteins were 5:1 for ENR-BSA.The antibodies showed the 50% inhibition of control values(IC50) of 60 ng/mL by ci-ELISA.The antigenic conjugates were artificially synthesized and the anti-enrofloxacin antibody received,which provides the experimental basis for the further study of ELISA or TRFIA.
