高强度聚焦超声波对体外细粒棘球蚴生发细胞生长增殖的影响
Effects of High Intensity Focused Ultrasound on the Growth of Hydatid Germinal Cells of Echinococcus Granulosus in Vitro
【摘要】 目的了解高强度聚焦超声波(HIFU)体外抑制细粒棘球蚴生发细胞生长增殖的作用。方法观测不同辐照剂量的HIFU对生发细胞的急性杀伤作用,获得剂量-效应关系;选择致10%生发细胞即刻死亡的剂量辐照后将细胞继续培养,噻唑蓝法、电镜、流式细胞仪等检测HIFU对生发细胞的生长抑制作用以及对细胞周期的影响和诱导凋亡作用。结果 HIFU对生发细胞有明显的即刻杀伤作用,20W×20s和40W×10s是致10%生发细胞即刻死亡的超声剂量。用此超声剂量作用后存活率为90%的生发细胞继续体外培养6,12和24h,各时点的细胞死亡率与对照组相比均具有显著的统计学差异(P<0.05);而照射后不同培养时点差异亦具有统计学意义(P<0.05),24h抑制作用表现最为明显;HIFU辐照后继续培养的细胞透射电镜下观察到凋亡改变,流式检测出辐照组凋亡峰,细胞周期S期细胞百分比升高,而G期降低。结论 HIFU能够即刻杀灭棘球蚴生发细胞并能抑制其在体外的生长,辐照后的细胞周期受阻于G2-M期,并能诱导生发细胞凋亡。
【Abstract】 Objective To evaluate the effect of high intensity focused ultrasound (HIFU) on the growth and pro liferation of germinal cells of echinococeus granulosus in vitro. Methods Germinal cells were irritated with different dosage of HIFU in vitro and the cytotoxic effect was investigated by trypan blue exclusion assay to obtain the dosageeffect curve. 25 W×20 s and 50 W×10 s were chosen as 10% lethal dosages of HIFU. The vitality rates of cells treated with 10% lethal dosages of HIFU were detected by MTT at 6,12 and 24 hours after HIFU irritation. Electron microscopy and flow cytometry were applied to observe apoptosis in cells and analyze cell cycle. Results Compared with the untreated group, the vitality of germinal cells significantly decreased immediately after treated by HIFU. The growth of the cells 90% surviving the exposure of HIFU was obviously suppressed. Typical morphological changes of apoptosis such as apoptotic body were found under electron microscope. The cell number increased in S phase and decreased in G phase with the increased apoptotic indices. Conclusions HIFU showed not only immediate-killing effect but also growth-inhibiting effect, as well as apoptosis-indueing effect on the hydatid germinal cells of echinococcus granulosus in vitro.
