聚焦超声辐照对血管瘤及血管畸形动物模型组织影响
Tissue Change of Animal Modle of Hemangioma and Vascular Malformation Irradiated by High-intensity Focused Ultrasound
【摘要】 目的以公鸡鸡冠模拟血管瘤及血管畸形,探讨聚焦超声治疗血管瘤及血管畸形的可行性及机理。方法 40只莱亨公鸡随机均分为A、B、C、D 4组,A、B、C组为实验组,D组为对照组,实验组使用CZP型超声波治疗仪以相同方法扫描式辐照鸡冠右侧,功率4.5W,频率9MHz,脉冲1000Hz,间距2mm,来回扫描5次共10min,观察辐照后鸡冠活体改变,3、7、14 d后分别取A、B、C组辐照区鸡冠组织,常规制片,HE染色后光镜观察其皮下血管层改变并与D组对照,计算10个高倍视野下血管平均密度并进行统计学分析。结果聚焦超声辐照后,辐照区鸡冠颜色变淡、增厚、变硬;光镜下不同程度水肿,部分血管闭塞,纤维组织增多,血管平均密度[A组(28.5±9.6),B组(30.5±11.8),C组(25.3±7.9)个/400倍HP]均比对照组[(56.8±13.7)个/400倍HP]显著减少(P<0.05),实验组之间血管平均密度无显著差异(P>0.05)。结论聚焦超声辐照可引起鸡冠血管闭塞及组织纤维化,可能是治疗皮肤、皮下血管瘤及血管畸形的有效方法之一。
【Abstract】 Objective To explore the feasibility and mechanism of high-intensity focused ultrasound in treating hemangioma and vascular malformation simulated by cockscomb. Methods 40 Laiheng cocks were divided randomly into experiment groups which were A, B, C groups and control group as D group. Each group contained 10 cocks. The right-side cockscombs from experiment groups were irradiated by scanning five times with 10 minutes back and forth using CZP ultrasonic therapeutic apparatus, with power 4.5 W, frequency 9 MHz, impulse 1 000 HZ, interval 2 mm. Except that the live cockscombs irradiated by ultrasonic were noted, cockscombs from A、B、C groups were respectively cut away after 3、7、14 days to make into routine slices stained by HE, which were observed the subcutaneous vascular regions under light microscope compared with D group. The average density of blood vessel selected randomly from 10 high power visual field was calculated and analyzed statistically. Results The cockscombs became thick and hard with colour fading after irradiated by focused ultrasound. Incoordinately dropsical tissue, some occlusive blood vessel and increasing fibre were found under light microscope. The average density of blood vessel of experment groups (A group 28.5±9.6, B group 30.5±11.8, C group 25.3±7.9/400HP) decreased remarkably compared with the control group (P<0.05), with no significant changes among experiment groups (P>0.05) . Conclusions Focused ultrasound irradiating can induce cockscomb blood vessel obstruction and tissue fibrosis, which can be one of the effective treatment to hemangioma and vascular malformation in skin as well as in subcutaneous stratum.
