乙酰谷酰胺对戊四氮致痫大鼠大脑病理改变以及GABABR和NMDAR1的影响

【摘要】 目的研究乙酰谷酰胺对戊四氮致痫大鼠大脑病理改变的影响,以及乙酰谷酰胺对大鼠脑内GABABR和NMDAR1表达的影响,寻找乙酰谷酰胺对癫痫的治疗的机制。方法戊四氮腹腔注射建立癫痫模型,分组给与不同剂量乙酰谷酰胺治疗,经主动脉灌注取脑组织,HE染色观察大脑病理改变情况,用免疫组织化学方法检测GABABR和NMDAR1的表达。结果各组神经细胞坏死率有显著差异(P<0.05),治疗组和模型组GABABR表达明显低于空白对照组(P<0.05),NMDAR1表达明显高于空白对照组(P<0.05)但各治疗组之间无明显差异(P>0.05)。结论乙酰谷酰胺对癫痫导致的脑损伤有治疗作用,并且其疗效呈现一定的剂量和治疗时间依赖性。乙酰谷酰胺对脑内GABABR和NMDAR1表达的影响不显著,因此推测乙酰谷酰胺的作用机制不是通过这两个受体起作用,具体机制有待进一步研究。

【Abstract】 Objective To investigate the effect of acetylglutamide on pathological changes,GABABR and NMDAR1 of rats with seizure induced by pentetrazole.Methods The rats were injected with PTZ into abdomen to induce seizure. Then those rats were subjected separately with different doses of acetylglutamide for different periods.The rats were perfused through aorta,and the brain tissues were fixed and embedded with paraffin later.The expression of GABABR and NMDAR1 in the hippocampus was measured by immunohistochemical method.Furthermore,the pathological changes of the brain were observed by microscope.Results Neuron death rate of each group was significantly different (P<0.05).The expression of GABABR in model group and treatment groups was significantly decreased compared with the control group (P<0.05),and that of NMDAR1 was significantly increased (P<0.05).But there was no obviously contrast among the model and treatment groups (P>0.05).Conclusion Acetylglutamide possesses the active effect on epilepsy,and its therapeutic effect correlates with its dosage and therapy time course to some extent. But the effect of acetylglutamide on the expression of GABABR and NMDAR1 is not obvious.So it is conferred that GABABR and NMDAR1 are not the channel through which acetylglutamide acts.We will do further researches to find the mechanism.
