蝉衣多糖的提取及理化性质测定
Extraction of Periostracum Cicadae Polysaccharide and Determination of its Physical and Chemical Property
【摘要】 目的:研究中药蝉衣中多糖的制备和性质。方法:将蝉衣洗去泥沙,在一定温度条件下采用酸、碱的方法,进行脱钙、脱蛋白、脱乙酰等处理,低温烘干,获得一定纯度的蝉衣多糖;采用酸碱滴定法测定脱乙酰度,凯氏定氮法测定含量,红外光谱法测定其部分性质,探讨了蝉衣多糖的溶解性和与部分试剂的反应。结果:蝉衣多糖的平均提取率为30.91%,脱乙酰度为96.95%,在酸性溶液中有很好的溶解度,与甲基红、甲基橙、苯胺蓝、蒽醌、荧光黄、萘酚等试剂反应呈现不同的颜色,其红外图谱与壳聚糖极为相似。结论:蝉衣中含有大量多糖,属碱性多糖,容易制备且纯度高,值得开发利用。
【Abstract】 Objective:To study the preparation and the physicochemical property of polysaccharide from periostracum cicadae.Methods: The periostracum cicadae was washed and dried to gain purified polysaccharide with acid and alkali methods and through the demineralized,deproteinized,and deacetylated processing at a certain temperature conditions.The acid-base titration was used to determine deacetylation degree;Kjeldahl method to determine its content;Infrared spectroscopy to determine other parts of its property for the solubility and the reaction with some reagents of the periostracum cicadae polysaccharide.Results: The average extraction rate of the periostracum cicadae polysaccharide was 30.91%,the deacetylation degree was 96.95%,with a good solubility in acidic solution,showing different colors with the reagents of methyl red,methyl orange,aniline blue,anthraquinone,fluorescent yellow and naphthol,etc.Its IR spectra were very similar with the chitosan.Conclusion: The periostracum cicadae polysaccharide has a large number of polysaccharide and alkaline polysaccharide and can easily be prepared and made highly purified.
