小檗碱在抑制兔颈动脉粥样硬化形成中的作用
The Mechanism of Berberine on Preventing Carotid Atherosclerosis in Rabbits
【摘要】 目的:探讨小檗碱抑制家兔颈动脉粥样硬化形成的作用机制。方法:24只雄性新西兰大白兔随机分为3组。正常对照组予普通饮食5周;模型组予高脂饲料喂养1周后,行右侧颈动脉内膜空气干燥术,再高脂喂养4周制作颈动脉粥样硬化模型;小檗碱处理组每日同时予颈动脉粥样硬化模型兔灌服小檗碱。观察颈动脉病理改变,并检测组织中巨噬细胞沉积和核因子-кB(NF-кB)、单核细胞趋化蛋白-1(MCP-1)表达情况。结果:模型组颈动脉有明显粥样硬化斑块形成,可见大量巨噬细胞沉积,NF-кB p65和MCP-1蛋白及mRNA表达明显(P<0.05),小檗碱组颈动脉硬化程度较轻,巨噬细胞明显减少,NF-кB p65和MCP-1蛋白及mRNA表达较模型组低(P<0.05),NF-кB与MCP-1二者呈正相关(r=0.958,P<0.01)。结论:小檗碱可能通过抑制NF-кB活性、降低MCP-1表达,减少巨噬细胞沉积而干预颈动脉粥样硬化形成。
【Abstract】 Objective To investigate the mechanism of berberine on preventing atherosclerosis(AS) in the carotid of rabbits.Methods Twenty-four New Zealand white rabbits were randomly divided into normal control group,carotid atherosclerosis model group and berberine group.The rabbits of control group were fed with normal diet.The model of carotid atherosclerosis was established with an air-drying operation on the right carotid and fed with a high cholesterol-diet in the second week for 4 weeks.The rabbits of berberine group were intragastricly administrated with berberine at a dose of 110 mg/kg daily from the first week for 5 weeks.The pathological change of right carotid arteries were observed with HE staining,the deposition of macrophage cells were assayed with immunohistochemistry staining,the expresssion of nuclear factor kappa B(NF-кB) mRNA and monocyte chemoattractant protein-1 mRNA were determined by RT-PCR.Results AS plaque and deposition of macrophage cells were obviously observed,the expression of NF-кB p65 mRNA and MCP-1 mRNA were increased in model group.In berberine group,the degree of carotid AS was slight,the number of macrophage cells was decreased,the expression of NF-кB p65 mRNA and MCP-1 mRNA was significantly lower than those of model group(P<0.05),meanwhile the expression of NF-кB p65 mRNA and MCP-1 mRNA was significantly correlated(r=0.958,P<0.01).Conclusion Berberine could inhibit the activity of NF-κB and expression of MCP-1 so as to decrease the deposition of macrophage cells,furthermore,the formation of carotid atherosclerosis was prevented.
