大鼠局灶脑缺血预处理对核因子-кB活化及神经元凋亡的影响

【摘要】 目的:观察局灶脑缺血预处理对核因子-кB(nuclear factor-кB,NF-кB)表达及神经元凋亡的影响。方法:将36只SD大鼠随机分为3组,对照组给予两次假手术,其余2组分别在2h大脑中动脉阻塞(MCAO)及22h再灌注前3d给予10min的预缺血或假手术,比较各组神经元凋亡及NF-кB的表达变化。结果:与缺血组相比,预缺血组凋亡神经元数减少(P<0.01),同时伴NF-кB表达降低(P<0.01)。结论:10min MCAO缺血可减少再次缺血后的神经元凋亡,抑制NF-кB激活,NF-кB活化受抑可能是局灶性脑缺血预处理抗凋亡的分子机理之一。

【Abstract】 Objective:To investigate the effect of focal cerebral ischemic preconditioning on activation of nuclear factor-ΚB(NF-кB) and neuronal apoptosis in rats.Methods:10 min MCAO was used as preconditioning(PC).36 SD rats were divided into 3 groups in which one group received twice sham surgery(SS) only,and the other two groups received 2 h middle cerebral artery occlusion(MCAO) followed by 22 h reperfusion with IPC or SS 3 d before respectively.Activation of NF-кB and neuronal apoptosis were evaluated in each group by immunohistochemistry and TUNEL staining.Results:Compared with the SS group,there were fewer apoptotic neuron and lower NF-кB immunoreactivity in PC group.Conclusion:10 min PC are strong enough to reduce ischmic neuronal apoptosis and inhibit activation of NF-кB.Inhibition of NF-кB may play an important role in protecting neuron against apoptosis.
